Optimizing lipase activity, enantioselectivity, and stability with medium engineering and immobilization for beta-blocker synthesis.

Lipase from Pseudomonas cepacia showed poor activity and moderate enantioselectivity (E) in pure aqueous systems for hydrolysis of a racemic mixture (+/-)-1-chloro-2-acetoxy-3-(1-naphthyloxy)-propane, which is a potential intermediate for beta-blocker synthesis. However, addition of polar organic solvents to the reaction can change both the activity and the enantioselectivity for this chiral reaction significantly. It was observed, in general, that the activity increases and the enantioselectivity decreases with the increase in the polarity of the organic solvent added to the medium. Among the six solvents chosen (i.e., dimethylsulfoxide [DMSO], 1, 4-dioxane, dimethylformamide [DMF], acetone, 1-propanol, and tetrahydrofuran [THF]), maximum activity and minimum enantioselectivity was obtained with DMSO, whereas minimum activity and maximum enantioselectivity was obtained with THF as the cosolvents. In the subsequent studies, native or polyethylene glycol (PEG)-modified lipase was immobilized by entrapping in Caalginate gel beads. In a fixed-bed continuous reactor containing these catalyst beads, the enzyme was found to be at least three times more enantioselective than the native form in a batch reactor. This fixed-bed reactor with the beads could be operated with high concentration of acetone (33% v/v) for about 1 month without a significant loss of enzyme activity and enantioselectivity.

New anionic polyelectrolyte hydrogel for corneal surgery.

A new high-water-content (78%) anionic polyelectrolyte hydrogel was obtained by phase inversion (demixion) of a polymer solution containing 9.0% poly(acrylonitrile sodium methallylsulphonate), 85.0% dimethylformamide, and 6.0% saline solution (0.9% NaCl). The hydrogel is permeable to water, saline, urea, creatinine, glucose, human albumin, and saline-dissolved oxygen. Investigation of the interactions between human serum and surfaces prepared with the new yielded hydrogel, compared to serum interaction with silica-free silicone (RTV), regenerated cellulose (Cuprophan), MMA/PVP copolymer (Lidofilcon), PMMA (Perspex), FIFE (Gore-Tex), and poly(acrylonitrile sodium methallylsulphonate) hemodialysis membrane (AN-69), showed the hydrogel and hemodialysis membrane (both prepared with AN-69 copolymer) to be the only materials devoid of complement (C')-activating ability.

Control of fibronectin receptor expression by fibronectin: antisense fibronectin RNA downmodulates the induction of fibronectin receptor by transforming growth factor beta1.

The results of our previous studies of mouse embryo fibroblasts showed that fibronectin expression and fibronectin receptor expression are tightly coregulated and that fibronectin modulates expression of its receptor in response to treatment with the differentiation-inducing agent N,N,-dimethylformamide (Varani and Chakrabarty, 1990, J. Cell. Physiol., 143:445-454; Huang et al., 1994, J. Cell. Physiol., 161:470-482). We also found that transforming growth factor beta1 (TGFbeta1) induces a more differentiated phenotype in the epithelium-derived human colon carcinoma cell line Moser and upregulates the expression of both fibronectin and its receptor (Huang and Chakrabarty, 1994, Int. J. Cancer, 57:742-746). By expressing antisense fibronectin RNA in Moser cells, we have downregulated fibronectin mRNA expression and thus blocked the ability of TGFbeta1 to induce fibronectin expression (Huang and Chakrabarty, 1994, J. Biol. Chem., 269:28764-28768). In this study, we examined the effect of antisense fibronectin RNA expression on the induction of fibronectin receptor by TGFbeta1 and tested the hypothesis that the induction of fibronectin expression by TGFbeta1 is required for the induction of fibronectin receptor expression. Blocking fibronectin induction by TGFbeta1 attenuated the ability of TGFbeta1 to upregulate the expression of cell-surface fibronectin receptors, alpha5beta1 integrin expression, and adhesion to extracellular matrix fibronectin. We therefore conclude that induction of fibronectin expression is required for optimal upregulation of fibronectin receptor expression by TGFbeta1.

Cryochemistry: freezing effect on peptide coupling in different organic solutions.

The freezing effect on peptide coupling in organic solutions of different polarity has been investigated and compared with the results obtained in liquid phase. The model reaction of DCC-activated coupling of Boc-Ala-Phe-OH with H-Ala-OBu(t) has been carried out in dioxane, dimethylsulfoxide and formamide, as well as in mixtures (90%/10%, v/v) of dioxane with acetonitrile, dimethylformamide, dimethylsulfoxide and formamide. The reactions have been traced and evaluated by RP-HPLC analysis. Freezing the reaction mixture resulted in all cases in a significant suppression of the N-dipeptidylurea side-product formation together with a slight decrease of tripeptide epimerization. The coupling yields and the side effects depended on the solvent, with the dioxane and dioxane/acetonitrile mixture produced the best results. The role of freezing and solvent in the improved results is discussed.

Nonaqueous ionic liquids: superior reaction media for the catalytic Heck-vinylation of chloroarenes

Nonaqueous ionic liquids, that is molten salts, constitute an activating and stabilizing noninnocent solvent for the palladium-catalyzed Heck-vinylation of all types of aryl halides. Especially with chloroarenes an improved activity and stability of almost any known catalyst system is observed as compared to conventional, molecular solvents (e.g. dimethylformamide (DMF), dimethylacetamide (DMAc), N-methyl-2-pyrrolidinone (NMP), or dioxane). Thus, even catalytic amounts of ligand-free PdCl2 yield stilbene from technically interesting chlorobenzene and styrene in high yield (turnover number (TON) = 18) without the need for further promoting salt additives such as tetraphenylphosphonium chloride. The scope of the new reaction medium is outlined for the first time for the vinylation of various aryl halides using different mono- and disubstituted olefins as well as a variety of known palladium(0) and palladium(II) catalyst systems. Furthermore, a novel means of catalyst recycling is presented and its scope is evaluated.

Aminopyrazine analogues as chemiluminescence derivatization reagents for pyruvic acid.

Aminopyrazine analogues were studied as sensitive and selective chemiluminescence derivatization reagents for pyruvic acid. These analogues reacted with pyruvic acid under acidic conditions at 100 degrees C to produce Cypridina luciferin derivatives, which exhibit chemiluminescence by reaction with hydrogen peroxide in the presence of potassium t-butoxide in dimethylformamide. Of the four aminopyrazine analogues (2-amino-5-phenylpyrazine, 2-amino-5-(4-hydroxyphenyl)pyrazine, 2-amino-5-(3,4, 5-trimethoxyphenyl)pyrazine, and 2-aminoquinoxaline), in the present test 2-amino-5-(3,4,5-trimethoxyphenyl) pyrazine was the most sensitive for pyruvic acid, and the chemiluminescence intensity was about four times higher than that obtained with aminopyrazine.

Lentivirus-mediated gene transfer of gp91phox corrects chronic granulomatous disease (CGD) phenotype in human X-CGD cells.

BACKGROUND: Chronic granulomatous diseases (CGD) are caused by impaired antimicrobial activity in phagocytes, due to the absence or malfunction of the respiratory burst NADPH oxidase. Two-thirds of the patients have mutations in their X-linked CGD gene encoding gp91phox, the largest subunit of the NADPH oxidase. METHODS: Aimed at gene therapy of X-CGD already at the level of resting pluripotent hematopoietic stem cells, we generated an advanced HIV-1-based vector with self-inactivating (SIN2) features containing the therapeutic gp91phox gene. In this vector an internal cytomegalovirus (CMV) promoter exclusively drives transgene expression. The green fluorescent protein (GFP) served as reporter for evaluation of gene transfer and expression in the human myeloid PLB985 X-CGD cell line. RESULTS: The X-CGD cells were efficiently transduced by the VSV-G pseudotyped lentivirus constructs (up to 74% GFP+ cells at 3 days post-transduction). CMV-driven GFP-expression was stable for at least 3 weeks after transduction and persisted after granulocytic differentiation of the target cells. Using the lentivector with the gp91phox transgene, 26% and 48% of the X-CGD cells expressed gp91phox at Days 2 and 20 after co-culture with 293T producer cells, respectively. Upon granulocytic differentiation of the transduced X-CGD cells with dimethylformamide (DMF), up to 63% (mean 49%, n = 7) of the cells were found to be functionally reconstituted with mean levels of superoxide production of 31% (n = 7) compared to wild-type PLB985 cells. CONCLUSION: Lentivirus vectors expressing gp91phox are able to at least partially correct human myeloid X-CGD cells.

Altering the inclusion properties of CTV through crystal engineering: CTV, carborane, and DMF supramolecular assemblies

The complexes [Na(CTV)2(OH)(H2O)](H2O)(DMF)2(o-carborane) (3; CTV = cyclotriveratrylene), [K(OH)(CTV)(DMF)]2(o-carborane) (4), [(DMF)(CTV)]2(H2O)4(o-carborane) (5), and (o-carborane)(CTV)(DMF)2 (6) all form as crystalline inclusion complexes from N,N'-dimethylformamide (DMF) solution. Complexes 3 and 4 are the first reported examples of CTV acting as a chelating ligand, with two CTV molecules coordinating cis to the six-coordinate M+ centers (M=Na, K). The extended structures of complexes 3-5 are similar, forming extended coordinate and/or hydrogen-bonding interactions and all feature intracavity complexation of DMF by CTV, while the complex 6 forms an assembly of (o-carborane) intersection of two sets (CTV) ball-and-socket supermolecules with DMF as a channel-type included guest.

Studies of the synthesis of SSZ-25 zeolite in a "mixed-template" system.

The synthesis of aluminosilicate zeolite, SSZ-25, is described using a two-component organic guest molecule strategy. This method has been recently described by us and is quite effective in crystallizing a variety of aluminosilicate zeolites with reduction in template cost. In this instance the original organic guest component used in the discovery of SSZ-25, N',N',N'-trimethyl-2-adamantammonium cation, is used in conjunction with a variety of smaller amines. The amine is the major organic component in the synthesis and the quaternary ammonium compound, while a minor component, is essential for structure direction to the desired zeolite product. Studies here show that the adamantyl component is preferentially taken up during crystal growth. Our studies showed that the use of the multi-organic component approach to synthesis resulted in a faster and cheaper route to crystallizing SSZ-25. The SSZ-25 has been described in the literature as having the MWW topology and as such has two different channel systems accessed by 10-ring openings; the channel systems do not intersect. One set of 10-rings open into large cavities. In this study we used NMR spectroscopy to examine the as-made materials and to see if the two organic components are used in different capacities in the synthesis, probing if the admantyl component is only found in these larger cavities. The crystalline products could be affected by solvent extraction with polar solvents like dimethylformamide: the X-ray powder patterns and elemental analyses changed with solvent treatment. The solvent-treated crystals were studied by NMR spectroscopy, elemental analysis, and argon adsorption. The two-component organic guest approach was also found to be quite flexible not only with regard to the amine, but also towards the quaternary ammonium compounds. Non-adamantyl polycyclic templates could be used, and polar but non-quaternized adamantyl derivatives also succeeded in directing towards SSZ-25 formation. On the other hand. in one instance it was shown that the two-component approach favors SSZ-25 as the kinetic product but at longer run times Ostwald ripening was observed, leading to the disappearance of SSZ-25 and formation of ferrierite zeolite and quartz.

Crucial influence of solvent and chirality-the formation of helices and three-dimensional nets by hydrogen-bonded biimidazolate complexes.

Deprotonation and recrystallisation of racemic [Co(2,2'-biimidazole)3][NO3]3 by ammonia in water/dimethylformamide solutions gave crystals of [Co(Hbiim)3] x 0.8H2O x 0.5DMF (2: Hbiim = monoanion of 2,2'-biimidazole, DMF = dimethylformamide), a porous material that contains fourfold interpenetrating (10.3) three-dimensional nets formed by neutral, hydrogen-bonded [Co(Hbiim)3] units, with DMF molecules in the narrow channels. Recrystallisation of [delta-Co(2,2'-biimidazolate)3] gave helices instead of the expected (10,3)-a net. These results are discussed in the light of additional density functional theory and molecular mechanics calculations and the X-ray structure of [Co(H2biim)3][NO3]3.

Solid-state transformations of zinc 1,4-benzenedicarboxylates mediated by hydrogen-bond-forming molecules.

The zinc 1,4-benzenedicarboxylates [Zn3(bdc)3(H2O)3] . 4 DMF (1; bdc = 1,4-benzenedicarboxylate), [Zn(bdc)(H2O)] . DMF (2), and [Zn(bdc)] DMF (3) crystallise at room temperature from mixtures of toluene/ dimethylformamide (DMF) under concentrated, dilute and dry conditions, respectively. The structure of phase 1 (monoclinic: P2(1)/c, a 13.065(1), b = 9.661(1), c = 18.456(1) A, beta = 106.868(2) degrees) consists of layers containing stacks of three zinc cations linked by mono- and bidentate bdc groups. Structure 1 converts to the known phase 2 by an irreversible, reconstructive phase transformation, whereas 2 and 3 interconvert reversibly upon the loss or addition of water. Removal of all solvent molecules included during crystallisation gives poorly crystalline [Zn(bdc)] (4), which is readily converted to highly crystalline solids upon contact with hydrogen-bond-forming molecules such as water, DMF and small alcohols. The crystal structures of the mono- and dihydrates [Zn(bdc)(H2O)] (6) and [Zn(bdc)-(H2O)2] (7) have been determined ab initio from powder X-ray diffraction data (compound 6, monoclinic: C2/c, a = 17.979(1), b = 6.352(1), c = 7.257(1) A, beta=91.477(1) compound 7, monoclinic: C2/c, a = 14.992(1), b = 5.0303(2), c = 12.098(1) A, beta = 103.82(1) degrees). The methanol adduct [Zn3(bdc)3] . 6CH3OH (5) is the same as that prepared previously by direct crystallisation. Comparison of these adduct structures with those prepared directly reveal that they are formed by in situ recrystallisations. Subsequent removal of included molecules gives amorphous [Zn(bdc)], which can be recrystallised again when placed in contact with hydrogen-bond-forming molecules.

Mechanism of the electrochemical conversion of aryl halides to arylzinc compounds by cobalt catalysis in DMF/pyridine.

The study of the electrochemical behavior of cobalt bromide, CoBr2, in the presence of zinc bromide, ZnBr2, and aryl halides, ArX, in a dimethylformamide (DMF)/pyridine (9:1, v/v) mixture allowed us to complete the study of the mechanism of the electrochemical conversion of aryl halides into arylzinc compounds by using cobalt catalysis. The last step of the catalytic process has been shown to be a transmetalation reaction between the arylcobalt(II) species and zinc ions that regenerates the cobalt(II) catalyst. The effect of zinc bromide on each step of the catalytic cycle has been studied. It is especially shown that the presence of ZnBr2 stabilizes the electrogenerated Co1 but has no effect on the rate constant of the oxidative addition of aryl halides, ArX, to Co1. Rate constants for the disproportionation reaction of Co1 and the oxidative addition have been determined in the presence of ZnBr2 and compared with the values obtained in its absence.

Host-guest chemistry of the chromium-wheel complex [Cr8F8(tBuCO2)16]: prediction of inclusion capabilities by using an electrostatic potential distribution determined by modeling synchrotron X-ray structure factors at 16 K.

The structure and detailed electron density distribution (EDD) of the large octanuclear chromium-wheel host complex [Cr8F8(tBuCO2)16] (1) has been determined from synchrotron X-ray structure factors collected at 16(5) K. The complex has a central cavity with a minimum entry distance between carbon atoms of the pivalate methyl groups (pivalic acid = tBuCO2H) of 4.027(4) A on one side of the molecule and 7.273(4) A on the other. The screened side of the molecule can be "opened" by rotation of methyl groups to create a strained host structure, which is compensated for by improved host-guest and host-solvent interaction. The EDD of the 272-atom complex (1144 e-) was determined by multipole modeling based on the experimental structure factors. 3d orbital populations on the Cr atoms and topological analysis of the EDD show that the covalent part of the metal-ligand interactions consists mainly of sigma donation from the ligands, but that overall the interactions are predominantly electrostatic. The electrostatic potential (EP) has been calculated from the experimental EDD. Knowledge of the geometry of the naked complex 1 as well as the EP in the central cavity of this molecule allows us to deduce which characteristic properties guest molecules must have to be accepted into the void. To probe these predictions, a series of complexes of 1 with different guest inclusions were synthesized (2 = 1 + N,N'-dimethylformamide (DMF), 3 = 1 + N,N'-dimethylacetamide (DMA), 4 = 1 + DMA + DMF, 5 = 1 + 2CH3CN), and their structures were examined by using X-ray diffraction data measured at 120(1) K. Results of these studies indicate that in the crystalline state, the optimal guest molecule should be linear and possess a permanent dipole. Attempts to crystallize the host complex with cations incorporated into the cavity were fruitless, although electrospray ionization mass spectrometry showed that a [1 + potassium]+ entity pre-exists in solution and can be transferred intact into the gas phase.

Highly stable and biocompatible nafion-based capsules with controlled permeability for low-molecular-weight species.

Biocompatible hollow capsules have been formed by electrostatic layer-by-layer self-assembly of a perfluorinated ionomer (Nafion) in alternation with ferric ions onto polystyrene latex particles or organic microcrystals, followed by dissolution of the cores by tetrahydrofuran or dimethylformamide. The stepwise growth of multilayers was followed by UV-visible spectroscopy and microelectrophoresis. The formation of hollow capsules was verified by confocal laser scanning microscopy and scanning force microscopy. The hollow Fe3+/Nafion capsules displayed high stability over a wide range of pH values and at high temperature. Fluorescein transport through the Fe3+/Nafion capsule wall was studied by means of photochemical bleaching and recovery (PBR) of the capsule interior. A diffusion model is suggested to calculate the diffusion coefficient for low-molecular-weight species, which was determined to be in the order of 10(-12) cm2s. The permeability can be manipulated by changing the wall thickness of the capsules.

Synthesis of some novel quinoline-3-carboxylic acids and pyrimidoquinoline derivatives as potential antimicrobial agents.

The synthesis and in vitro antimicrobial evaluation of several quinoline and pyrimidoquinoline derivatives are described. Treatment of 7-substituted quinolin-2(1H)-one-3-carboxylic acids 2a-c with phosphoryl chloride or thionyl chloride gave rise to the 7-substituted 2-chloroquinoline-3-carboxylic acids 3a-c and 7-substituted 2-chloro-3-chlorocarbonylquinolines 5a-c respectively. The 2-chloro function in compounds 3a-c was replaced by 2-aminothiazole or 2-aminopyridine to give 2-(thiazol-2-yl)aminoquinoline-3-carboxylic acids 4a-c or 2-(pyrid-2-yl)aminoquinoline-3-carboxylic acids 4d-f. Treatment of 5a-c with the same heterocyclic amines at room temperature furnished the corresponding 7-substituted 2-chloro-3-heteryl-aminocarbonylquinolines 6a-f. The tetracyclic 9-substituted thiazolo[3', 2':1, 2]-pyrimido[4, 5-b]quinolin-5-ones 7a-c and 10-substituted pyrido[1', 2':1, 2]-pyrimido[4, 5-b]quinolin-6-ones 7d-f were synthesized by heating 5a-c with the heterocyclic amines in toluene or by heating 6a-f under reflux in dimethylformamide. The products were evaluated in vitro for potential antimicrobial activity.

Electrogenerated chemiluminescence from violanthrone.

Violanthrone, an emitter of exceptionally bright chemiluminescence, was examined in dimethylformamide solution to determine whether it also emits particularly bright electrogenerated chemiluminescence (ECL). The ECL measurements were carried out using a cycled potential which was applied to platinum electrodes. At the maximum sweep rate of 80 V s-1 available, the intensity of the violanthrone ECL was still increasing with sweep rate and was c. 56% of that from rubrene, a bright, commonly used emitter of ECL. Furthermore, assuming that the emission arises from radical anion-cation recombination, the sweep rate dependence showed that the least stable radical ion (probably the cation) decays with a half-life shorter than 0.2 s.

Peptide-bond modification for metal coordination: peptides containing two hydroxamate groups.

Peptide-bond modification via N-hydroxylation has been explored as a strategy for metal coordination to induce conformational rigidity and orient side chains for specific molecular recognition. N-Hydroxyamides were prepared by reacting N-benzyloxyamino acid esters or amides with Fmoc-AA-Cl/AgCN (Fmoc: 9-fluorenylmethoxycarbonyl; AA: amino acid) in toluene or Fmoc-AA/HATU/DIEA in DMF (HATU: O-(7-azabenzotriazol-lyl)-1,1,3,3-tetramethyluronium hexafluorophosphate; DIEA: N,N-diisopropylethylamine; DMF: N,N-dimethylformamide), followed by deblocking of benzyl protecting groups. Novel linear and cyclic N,N'-dihydroxypeptides were efficiently assembled using Fmoc chemistry in solution and/or on a solid support. As screened by electrospray ionization-mass spectroscopy (ESI-MS), high iron-binding selectivity and affinity were attainable. Compounds having a spacer of two alpha-amino acids between the amino acids bearing the two hydroxamates, i.e., a spacer of 8 atoms, generated 1:1 iron complex species in the gas phase. Moreover, high performance liquid chromatography (HPLC), uv/vis, and (1)H-NMR analyses provided direct evidence for complex formations in solution. Significantly, the representative compound cyclo(Leu-Psi[CON(OH)]-Phe-Ala-Pro)(2) (P8) may serve as a robust metal-binding scaffold in construction of a metal-binding library for versatile metal-mediated molecular recognition.

Electrostatics dominate quadruplex stability.

Stabilization of nucleic acid structures results from a balance of multiple interactions, including electrostatics, base stacking, hydrophobic interactions, hydrogen bonding, van der Waals forces, etc. Nucleic acid quadruplexes are unusual structures in that their formation is driven by specific binding of metal ions. This unique mode of metal binding, which is tightly coupled to oligonucleotide folding, can engender correspondingly unique solution behavior. In particular, we show that addition of many cosolvents, such as primary aliphatic alcohols, increases the thermal stability of quadruplexes, as determined by melting temperature, Tm, in direct contrast to the response of duplexes to the same admixture of solvents. Thermal stability is observed to increase as the dielectric constant of the composite solvent decreases. This behavior suggests a dominant role for electrostatics in quadruplex formation and stability. Additional studies done with other cosolvents and solutes suggest that, in some cases, other forces may come into play, including the possibility of direct interaction with the quadruplex structure. Nonetheless, many cosolvents and small molecules, such as ethanol, dimethylformamide, and betaine, stabilize the quadruplex conformation in sharp distinction to their destabilization of DNA duplexes.
