Carbodiimide-mediated condensation of DNA onto microwells is investigated. DNA is bound onto the microwells by formation of a phosphoramidate bond between the 5' terminal phosphate group and the microwells. Immobilization of 25 to 30 ng DNA per well is obtained. DNA molecules bound covalently at only the 5' end are, ideally, perfect for hybridization. The practicability of DNA molecules bound to microwells for hybridization is investigated.

Mouse spleen cells could be preparatively separated into immunoglobulin positive (Ig+) and immunoglobulin-netative (Ig-)populations by incubating as many as 2 X 10(8) cells per 100 mm diameter petri plate coated with specifically purified goat anti-mouse immunoglobulin. The non-adherent population was 95% or more Ig-, and possessed graft versus host and cytotoxic effector activities, as would be expected for T cells. They could also give a mixed lymphocyte reaction and generate cytotoxic effector activity on culture in vitro. The adherent cells could not be released undamaged from plates coated with undiluted anti-Ig, but they could be released from plates coated with a 1/4 or 1/10 dilution of anti-Ig in an irrelevant antibody. The released cells were over 90% viable by trypan-blue staining, and 94% or more of the viable cells were Ig+.
Three variations of a new immunoassay which used physicochemically stable latex particles (LP) are described. New features include (1) separation of free and agglutinated LP by centrifugation after the addition of sucrose, and (2) combined use of two or three types of LP. Assay results for anti-rabbit IgG (anti-RG), human chorionic gonadotrophin (HCG), C3 components and insulin indicated excellence of these methods in sensitivity, reproducibility and preservability of reagents.
Horseradish peroxidase conjugates are inactivated by reaction with plastic enzyme immunoassay reaction vessels (Gilford Cuvette Paks). The loss of enzyme activity is prevented by Tween.
Qualitative and quantitative changes in the antigenic mosaic of coated solid-phase matrices occurring during storage may have a pronounced effect on the comparability of results obtained by enzyme immunoassay (EIA). We have used, as a model antigen for studying the effects of storage, a sonic extract of Borrelia burgdorferi and a totally automatic EIA procedure. The IgM antibody concentrations of the sera originally determined to be high decreased by almost one half during 1 week's storage of the solid phase of 4 degrees C. In contrast, the IgM values of other sera, originally measured at medium or low, doubled upon storage. Thus, after storage the rank order of the sera was markedly affected compared to the values obtained with antigen freshly applied to the wells. In parallel determinations of IgG antibody levels the EIU relative unitage values of the test sera generally remained constant upon storage. The only exception was the serum with the highest original antibody concentration. Its relative antibody unitage more than tripled over the storage time. It is recommended that the coating of solid-phase matrices should be done immediately before each run in all EIAs where microbial extracts are used as antigens.
