An evaluation of new processing protocols for in vivo NMR spectroscopy.

In vivo NMR spectroscopy is often complicated with problems of low signal-to-noise, poor resolution, undefined peak shapes, and nonlinear baselines despite the efforts of investigators to optimize their experiments. Several data processing options are available to spectroscopists to enhance resolution and signal-to-noise and/or to flatten baselines. There is some question about how these processing protocols affect quantitative information. This paper evaluates five different processing protocols for their ability to extract quantitative information from a set of nonideal spectra. Three of the protocols involve recently developed statistical signal processing methods, maximum entropy Fourier spectral deconvolution, linear prediction singular value decomposition, and baseline deconvolution. These protocols are compared with the conventional processing methods of convolution difference and zeroing initial data points of the FID. The methods are evaluated by use of a quantitative 31P model sample and also are demonstrated on surface coil 31P data.

Application of a 3D volume 19F MR imaging protocol for mapping oxygen tension (pO2) in perfluorocarbons at low field.

A limited flip angle gradient-echo 3D volume acquisition imaging protocol for mapping partial pressure of oxygen (pO2) in perfluorocarbon compounds (PFCs) at low field (0.14 T) is presented. The PO2 measurement method is based on the paramagnetic effect of dissolved molecular oxygen (O2) which reduces the PFC 19F T1. Specific objectives related to imaging of PFCs through use of the protocol include improved image signal-to-noise characteristics and elimination of 19F chemical shift artifacts. A parametric Wiener deconvolution filtering algorithm is used for suppression of 19F chemical shift artifacts. Application of the protocol is illustrated in a series of calculated PO2 maps of a gas equilibrated, multi-chamber phantom containing perfluorotributylamine (FC-43). The utility of the protocol is demonstrated in vivo through images of a commercially available perfluorocarbon based blood substitute emulsion containing FC-43 sequestered in the liver and spleen of a rat.

Screening for aneuploidies of ten different chromosomes in two rounds of FISH: a short and reliable protocol.

OBJECTIVE: To develop a DNA labelling protocol for the simultaneous detection of five different fluorescent chromosomal DNA probes within one round of hybridisation. In combination with a commercial five-colour probemix for the second round of hybridisation, this results in a fast and reliable Fluorescence in situ Hybridization (FISH) protocol, enabling the detection of 10 chromosomes within a working day. This is especially of use for Preimplantation Genetic Screening (PGS), when only single interphase nuclei are available for analysis and when time is restricted. METHOD: DNA probes were labelled with four different fluorochromes (Pacific Blue, Alexa Fluor 350, Alexa Fluor 594 and Alexa Fluor 488) using an ARES labelling kit, based on a two-step method. Aminoallyl-dUTPs were incorporated by nick translation, followed by chemical linking of the amino-modified fluorescent dye. The fifth colour was achieved by using two fluorescent dyes in the chemical reaction, resulting in dual labelling of the DNA probe and a fluorescence detectable with a specific filter set. These five probes were simultaneously hybridised in a first FISH round, followed by a second hybridisation with a commercial five-colour probemix, thus allowing the detection of chromosomes 1, 7, 13, 15, 16, 18, 21, 22, X and Y. The fixation and pre-treatment procedures of the blastomere nuclei were further optimised. RESULTS: Using this labelling and FISH protocol, probe hybridisation efficiency, when tested on lymphocyte nuclei, is 95 to 99%. With the fixation protocol, blastomere nuclei maintain good morphology and show condensed and clear signals even after the second round of hybridisation. CONCLUSION: This labelling method in combination with specific epifluorescence filters, enables an independent detection of five different chromosomes in one round of FISH. The whole process of biopsy, fixation and two rounds of hybridisation with the analysis of ten chromosomes can be completed within a day.

Therapeutic Exercise Protocol During Hospitalization in Pediatric Oncohematological Patients: Randomized Clinical Trial.

BACKGROUND: Leukemias, lymphomas, and central nervous system tumors are among the most common pediatric cancers and may lead to motor deficits, impaired balance, reduced muscle strength, fatigue, and decreased functional capacity. Early physiotherapy during hospitalization may help prevent inactivity and support functional preservation in this population. OBJECTIVE: To evaluate the effects of a therapeutic exercise program on quality of life, muscle strength, fatigue, and functional capacity in hospitalized pediatric oncohematological patients. METHODS: Thirty participants aged 8-17 years with oncohematological diseases were randomized to an intervention group (IG) or a minimal active physiotherapy comparator group (CG). Assessments included the 6-min walk test, handgrip dynamometry, the PedsQL Multidimensional Fatigue Scale, and the PedsQL Cancer Module at admission and discharge. The IG performed daily 25-min supervised sessions including aerobic, resistance, and breathing exercises with ambulation guidance, whereas the CG received breathing exercises and ambulation guidance. RESULTS: No significant group × time interactions were observed for total fatigue or its domains, overall quality of life or its assessed domains, handgrip strength, or six-minute walk test distance. Time-related changes were observed for some outcomes, but these occurred without evidence of differential change between groups and were not interpreted as effects of the structured exercise protocol. No intervention-related adverse events requiring permanent protocol discontinuation were recorded. CONCLUSION: The structured in-hospital therapeutic exercise protocol could be delivered under close clinical supervision without recorded intervention-related adverse events requiring permanent discontinuation. However, the structured protocol did not demonstrate superiority over the minimal active physiotherapy comparator for fatigue, quality of life, muscle strength, or functional capacity. These findings should be interpreted cautiously because of the small sample size, clinical heterogeneity, variable intervention exposure, and limited intervention-fidelity data. Statistical comparison of two handwashing protocols.

This paper describes statistical procedures for use in an experiment that compares two handwashing protocols. The evaluation of a handwashing protocol entails collection of the wash effluent. Colony counts for the effluent reflect the number of flora removed by the wash protocol. The analysis aims to formulate and estimate an odds ratio that represents the relative efficacy of a test as compared with a standard protocol.

Evaluation of the OECD 421 reproductive toxicity screening test protocol using 1-(butylcarbamoyl)-2-benzimidazolecarbamate (benomyl).

The OECD 421 reproductive toxicity screening test protocol was evaluated using the fungicide benomyl as a test compound at 10, 30, or 90 mg/kg per day. Male rats showed dose-dependent testicular degeneration after 28 days exposure, as expected on the basis of literature data. Dams in the high dose group, exposed from 14 days premating to postnatal day 6, had pups with decreased weights at postnatal days 1 and 6. Prenatal deaths were increased, but no malformations were found. In contrast, in a developmental toxicity test with exposure between gestation days 6 and 15, 90 mg/kg induced a high level of postimplantation loss, with ophthalmic malformations in the surviving offspring, in agreement with literature data. It is suggested that premating exposure in the OECD 421 protocol may have induced tolerance to the compound, e.g., by modulation of biotransformation in the dam. These findings indicate that the teratogenic potential of a compound need not necessarily be revealed using this screening test. The OECD 421 protocol appeared sufficiently sensitive to reveal the reproductive hazard of benomyl on the basis of prenatal deaths and testicular and pup weight effects. However, the absence of congenital anomalies in the offspring after benomyl treatment according to the OECD 421 protocol underscores the notion that lack of biological activity in the test should not be regarded as evidence of lack of activity of a compound on reproductive parameters.

The effects of variation of cryopreservation protocols on the immunogenicity of allogeneic skin grafts.

The use of allografts to effect wound closure on full thickness skin injuries is limited by unpredictable graft rejection times. If the period of graft take could be extended, the use of allografts would reduce the morbidity and mortality associated with these wounds. This study evaluated the effects of variation in the cryopreservation protocol on the viability and immunogenicity of skin using a murine model system. Immunogenicity was assessed by the stimulatory activity of C3H (H-2K) skin-derived epidermal cells (EC) in primary one-way EC/lymphocyte reactions with BALB/c (H-2d) and CBA (H-2K) responder lymphocytes. Viability was determined by measuring tetrazolium reductase activity. The following cryopreservation protocols were assessed: freezing at 1, 30, 64, and > 100 degrees C/min in 10 and 15% (v/v) Me2SO and freezing at 30 degrees C/min in 5 to 20% (v/v) Me2SO or glycerol. A cryopreservation protocol of 30 degrees C/min in 15% (v/v) Me2SO proved optimal for murine skin allograft storage and immunomodulation. The viability of skin treated by this protocol was maintained (78% of fresh skin viability, no significant difference analysis of variance). The stimulatory capacity of treated EC for H-2K and H-2d lymphocytes was 5 +/- 4 and 5 +/- 9% (+/- 95% confidence limits) of fresh EC (100%) activity. Langerhans cell numbers in epidermal sheets and EC suspensions did not correlate with the stimulatory capacity of fresh and treated EC for allogeneic lymphocytes. A functional impairment of Langerhans cell immunostimulatory capacity was implied.(ABSTRACT TRUNCATED AT 250 WORDS)

The power of the European Union protocol to test for carcinogenicity of inhaled fibers.

We evaluate the power of a recent protocol proposed by the European Union (EU) for testing the carcinogenicity of inhaled fibers. We assume that every fiber has oncogenic potential determined by its biopersistence. We use a recently estimated potency for the oncogenic potential of fibers together with experimentally determined "weighted" half-lives (WHL) of a dozen fibers to generate simulations of long-term bioassays conducted according to the EU protocol. We analyze these experiments using standard statistical techniques and determine the number of tests that would have yielded significant results. We conclude that the EU protocol will readily detect the carcinogenic effect of long-lived fibers, such as amosite with a WHL of more than 450 days, and usually detect the effect of fibers, such as RCF1a, with WHL of about 40-50 days. However, the EU protocol has very low power to detect effects of short-lived fibers, such as X607, with WHL of about 10 days.

Hospital Protocols and Policies that may Delay Early Identification and Thrombolytic Therapy of Acute Myocardial Infarction Patients.

Despite the compelling relationship between early treatment and outcome from reperfusion therapy in patients with acute myocardial infarction, significant delays in early treatment are imposed by the patient, prehospital systems, and hospital processes and protocols used in the identification and treatment of patients with myocardial infarction. A survey instrument designed to determine the prevalence of hospital policies and protocols that might delay or expedite treatment with thrombolytic therapy in patients with acute myocardial infarction was completed by 524 hospital participating in the National Registry for Myocardial Infarction (NRMI). Participating hospitals had treated 17,646 patients with tissue plasminogen activator. The door to drug time for the entire population of patients treated at each hospital was available. Door to drug times were compared between those hospitals that had a positive response to a policy and those that had a negative response to that policy. Among respondent hospitals, thrombolysis was excluded by protocol in 34.4% for age above 75 and in 55% for presentation after 6 hours of chest pain onset. Furthermore, 29.4% of hospitals required routine laboratory testing other than electrocardiography (ECG), including chest x-ray, prior to determination of eligibility for thrombolysis. Door to drug times were shorter in those hospitals with prehospital 12-lead ECG availability, assessment of the 12-lead ECG by the emergency department nurse and physician as soon as it was available, and initiation of thrombolysis by the emergency physician (in patients with clear-cut ST elevation myocardial infarction) without bedside cardiology consultation. Door to drug times were longer in those hospitals in which predecision laboratory results were required, written informed consent was mandated, and drug was initiated in the cardiac intensive care unit rather than in the emergency department itself. Door to drug times were not significantly different in those hospitals with a designated chest pain center compared with those operating under a focused patient care protocol. We conclude that the earliest possible hospital treatment of acute myocardial infarction patients may be precluded by multiple components of emergency department policies and process, many of them inappropriate for safe, efficient, and effective identification and management of these patients.

A computer-based protocol for semiautomated assignments and 3D structure determination of proteins.

A strategy is presented for the semiautomated assignment and 3D structure determination of proteins from heteronuclear multidimensional Nuclear Magnetic Resonance (NMR) data. This approach involves the computer-based assignment of the NMR signals, identification of distance restraints from nuclear Overhauser effects, and generation of 3D structures by using the NMR-derived restraints. The protocol is described in detail and illustrated on a resonance assignment and structure determination of the FK506 binding protein (FKBP, 107 amino acids) complexed to the immunosuppressant, ascomycin. The 3D structures produced from this automated protocol attained backbone and heavy atom rmsd of 1.17 and 1.69 A, respectively. Although more highly resolved structures of the complex have been obtained by standard interpretation of NMR data (Meadows et al. (1993) Biochemistry, 32, 754-765), the structures generated with this automated protocol required minimal manual intervention during the spectral assignment and 3D structure calculations stages. Thus, the protocol may yield an approximate order of magnitude reduction in the time required for the generation of 3D structures of proteins from NMR data.

Selection of ovarian stimulation protocol is related to IVF treatment outcome in women 35 years of age and older.

PURPOSE: The aim of this study was to assess if the woman's age influenced IVF treatment outcome when a long GnRHa-hMG or a CC-hMG ovarian stimulation protocol was used. Two hundred women were included in the study, 100 women under the age of 35 and 100 women 35 years of age and older (mean 31.8 years and 36.7 years respectively). In the younger group as well as in the older group 50 women were stimulated according to a GnRHa-hMG protocol and 50 women received a CC-hMG regimen. RESULTS: Significant differences between stimulation protocols were found in the older group for the mean numbers of oocytes recovered (4.7 vs 3.0), preembryos obtained (3.2 vs 2.0) and replaced (2.3 vs 1.7), as well as pregnancy (30% vs 10%) and delivery (24% vs 4%) rates per replacement. CONCLUSION: It is concluded that women over 35 years of age seem to have a more favorable outcome of IVF treatment when using a long GnRHa-hMG protocol compared with CC-hMG, while this difference was not as obvious and lacking statistical significance under the age of 35.

The relationship between test protocol and the development of exercise-induced hypoxemia (EIH) in highly trained athletes.

Healthy male endurance-trained cyclists [n = 11, age = 27.3 (3.9) years; mass = 73.0 (9.3) kg; height = 180.5 (6.9) cm; maximal oxygen consumption (VO2max) = 71.1 (5.8) ml.kg-1.min-1, mean +/- (SD)] were recruited to assess the relationship between test protocol and the development of desaturation of arterial hemoglobin with oxygen, during incremental exercise tests to maximal aerobic capacity (VO2max). All subjects demonstrated resting pulmonary function within normal limits [forced vital capacity (FVC) = 6.0 (0.9); forced expiratory volume (FEV1.0) = 4.9 (0.6); FEV1.0/FVC = 0.8 (0.1)] and completed three ramped VO2max tests (Mijnhardt KEM-3 electronically braked cycle ergometer) beginning at 0 W with increments of either 20,30 or 40 W.min-1. All periods of testing were separated by a minimum of 72 h. VO2max, peak minute ventilation (VEpeak) (Medical Graphics, CPX-D), peak heart rate (fcpeak), peak power output (Wpeak), and minimum percentage arterial oxyhemoglobin saturation (% SaO2min) (Omeda Biox 3740 pulse oximeter) were determined. There were no significant differences (p > 0.05) in VEpeak [191.5 (26.2), 196.0 (24.4), 194.3 (23.9) l.min-1] fcpeak [191.4 (7.0), 190.3 (5.5), 187.8 (5.9) beats.min-1], VO2max [5.0 (0.5), 5.1 (0.4), 5.1 (0.5) l.min-1] or %SaO2min [89.5 (1.5), 89.6 (1.3), 90.0 (2.3)] between protocols. The 20-W protocol [417 (27) W] demonstrated significantly lower Wpeak (P < 0.05) than the 30-W [434 (36) W] and 40-W [453 (38) W] protocols, indicating that peripheral fatigue may play an important factor in response to these tests. The results of this study demonstrate that arterial desaturation occurs as a result of intense exercise in highly trained athletes independent of the rate of attainment of VO2max.

Quality of life in patients with locally advanced head and neck cancer treated with chemoradiotherapy. Comparison of two protocols using the EORTC questionnaires (QLQ-C30, H and N35).

INTRODUCTION: The objective of this study is to assess the quality of life (QoL) of two groups of patients during treatment for locally advanced head and neck (H and N) cancer. MATERIAL AND METHODS: Two samples of 30 patients each in AJCC stages III and IV undergoing either of two chemo-radiotherapy protocols completed the EORTC QLQ-C30 general questionnaire and the QLQ-H and N35 H and N module on three occasions during the treatment and follow-up periods. We also collected clinical data. The QoL scores and their evolution over the three measurements were calculated and both protocols were compared during the treatment period. RESULTS: The QoL scores are acceptable in general. Limitations were observed in relation to toxicity, psycho-social and some functional areas during the treatment. QoL improved in the follow-up period. The clinical and QoL data are better in one of the two treatment protocols. DISCUSSION: The QoL scores indicate that the condition of the patients receiving the protocols was acceptable, considering the severity of their disease. The treatments were reasonably-well tolerated.

A cost-effective plasmid purification protocol suitable for fluorescent automated DNA sequencing.

A cost-effective, reliable, and reproducible method has been developed to produce good-quality, double-stranded plasmid DNA for automated sequence analysis. The method incorporates modifications to a previously described plasmid-purification protocol used in manual sequencing. The quality of the DNA produced from the present protocol is suitable for automated fluorescent sequencing. Using a dye-terminator sequencing protocol, most runs using plasmid DNA prepared using this protocol produced over 700 bases with greater than 99% base-calling accuracy.

Choosing a thallium-201 or technetium 99m sestamibi imaging protocol.

During the past decade progress in the field of myocardial perfusion imaging has resulted in a myriad of choices for perfusion imaging protocols, including choices in types of stressor, imaging modality, perfusion tracer, method of analysis, and a wide range of choices of imaging protocols. This review will examine this latter aspect in the context of choices of perfusion tracers that are now available for wide use. Manipulations of thallium protocols have revolved primarily around optimizing identification of stress defect reversibility and regional myocardial viability. The high image quality and long retention time afforded by the technetium-labeled agents allow the potential for streamlining imaging protocols to maximize efficiency and cost-effectiveness, such as performing stress only imaging in a subset of patients and performing simultaneous assessment of perfusion and function.

Effects of non-specific vs individualized exercise training protocols on aerobic, anaerobic and strength performance in severely obese subjects during a short-term body mass reduction program.

The purpose of the present study was to compare aerobic, anaerobic and strength performance changes induced by two short-term (3-week) body mass reduction programs based on the same low-calory diet (1200-1500 kcal/day), nutritional education and psychological counseling, but entailing different exercise training protocols. An individualized, low-volume and moderate-intensity exercise training (IET) was contrasted with a non-specific, high-volume, low-intensity exercise training (NET). Thirty obese in-patients (12 males, 18 females; mean age +/- SD: 33.9 +/- 9.4 yr, range: 19-51yr; mean BMI: 40.5 +/- 3.8 kg/m2, range: 35.3-51.4 kg/m2) were randomly divided in two gender-matched groups of 15 subjects each undergoing a different exercise training protocol. Maximum oxygen uptake (VO2max) determined with a submaximal indirect test on a bicycle ergometer, lower limb maximum power output (W(max)) determined with the jumping method, global motor capabilities determined by analysis of locomotor pattern during a short (8 m) running, maximum strength (1-RM) of upper and lower limb muscle groups determined with isotonic machines were tested before and after the program. Adherence to an individual exercise activity and maintenance of body weight (bw) loss were evaluated with a telephonic interview 6 months after the completion of the program. In both groups a significant (p < 0.001) and comparable weight loss was observed (IET: -4.27%; NET: -4.17%). In both groups VO2max and W(max) increased significantly (p < 0.05-0.001) when expressed relatively to body mass, while in absolute terms they were significantly (p < 0.001) improved only in IET group. 1-RM in all tested muscle groups was significantly increased in both IET and NET subjects (p < 0.001-0.01), but improvements were significantly greater in IET as compared with NET (p < 0.05-0.001). The analysis of locomotor pattern during the short running indicated that IET subjects significantly improved their global motor capabilities (p < 0.05-0.001), while no change was observed in NET group. After 6 months, IET subjects reported a level of spontaneously chosen physical activity significantly higher (p < 0.05) than NET subjects, displaying a trend of further decrease in bw. It was concluded that, although no difference in bw loss was appreciated between the two studied groups and significant improvements were found also in subjects performing NET protocol, the IET protocol offers better overall results in terms of muscle performance and physical fitness, with a possibly stronger motivation to subsequent exercise activity.

Feasibility and potential gains of enhancing the subacute rat study protocol (OECD test guideline no. 407) by additional parameters selected to determine endocrine modulation. A pre-validation study to determine endocrine-mediated effects of the antiandrogenic drug flutamide.

Groups of five male and five female Wistar rats were treated by gavage with 0, 1, 10, and 100 mg flutamide/kg body weight for at least 28 days to investigate whether proposed enhancements to the current subacute rodent OECD test guideline no. 407 could be included into the testing routine, which of the current and/or additional parameters would detect endocrine-mediated effects of flutamide reliably and sensitively, and to provide information on intra-laboratory variability. Two identical studies were performed concurrently. The enhanced protocol requests the additional determination of the specific hormones triiodothyronine, thyroxine, thyroid stimulating hormone, follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol, prolactin, testosterone, corticosterone; of oestrus cyclicity and necropsy of all females in the dioestrus stage; of the number of homogenization-resistant testicular spermatids and the number, motility, viability, and morphology of cauda epididymal spermatozoa; of additional organ weights (pituitary, ovaries, uterus, thyroid, male accessory reproductive organs); and of the histopathology of additional organs (pituitary, epididymides, coagulation glands, pancreas, vagina). From a technical standpoint, it was possible to conduct a study according to the enhanced protocol, however, with substantial additional effort, an increase in costs by some 67%, and logistic problems. In line with the specific pharmacological effect of flutamide, treatment-related changes were mainly found in male rats, while females were hardly affected by 100 mg/kg. In male rats, 100 mg/kg was the maximal tolerated dose resulting in reduced body weight gain, but no or little other effects on clinical, haematological, clinico-chemical, or behavioral parameters, and 1 mg/kg was the no-observed-adverse-effect level. Antagonism of peripheral androgen receptors by flutamide resulted in decreased relative organ weights of male accessory reproductive organs, changes that were reliably detected in both studies at 100 mg/kg, but only in one of both studies at 10 mg/kg. Corresponding histopathological changes were also detected reliably at 100 mg/kg. Antagonism of central androgen receptors by flutamide increased LH and FSH levels. LH stimulation of testicular Leydig cells in turn increased testosterone and estradiol levels. Again, all these changes were detected reliably at 100 mg/kg, but only in one of both studies at 10 mg/kg. Corresponding histopathological alterations (increase of LH- and FSH-secreting cells, Leydig cell hypertrophy) were detected reliably and sensitively at 10 mg/kg. Studies on liver enzymes performed outside the scope of the enhanced protocol showed that flutamide at 100 mg/kg generally induced hepatic enzyme activities, but decreased the activity of the sex-specific testosterone-dependent liver enzyme CYP2C11 in male rats. The laboratory methods employed yielded reliable results, i.e., 93.6% of the quantitative measurements obtained in both studies were in agreement. Doubling the animal number from five to ten per sex and dose does not increase the sensitivity of detection of endocrine-mediated effects above the level already provided by histopathological examination of groups of five animals. Some of the proposed enhancements evaluated (additional organgravimetry and histopathology) were helpful in detecting the endocrine-mediated effects of flutamide reliably, while others did not contribute towards this aim (spermatology resulted in doubtful effects, female cyclicity was not affected, hormone determinations provided mechanistic information). Ongoing testing according to the revised version of the enhanced OECD test guideline no. 407 protocol and using ten compounds interfering with the endocrine system by different mechanisms will result in the identification of the most appropriate enhancements.

Impairments of hippocampal synaptic plasticity induced by aggregated beta-amyloid (25-35) are dependent on stimulation-protocol and genetic background.

The aggregation of beta-amyloid to plaques in the brain is one of the hallmarks of Alzheimer disease (AD). Numerous studies have tried to elucidate to what degree amyloid peptides play a role in the neurodegenerative developments seen in AD. While most studies report an effect of amyloid on neural activity and cognitive abilities of rodents, there have been many inconsistencies in the results. This study investigated to what degree the different genetic backgrounds affect the outcome of beta-amyloid fragment (25-35) on synaptic plasticity in vivo in the rat hippocampus. Two strains, Wistar and Lister hooded rats, were tested. In addition, the effects of a strong (600 stimuli) and a weak stimulation protocol (100 stimuli) on impairments of LTP were analysed. Furthermore, since the state of amyloid aggregation appears to play a role in the induction of toxic processes, it was tested by dual polarisation interferometry to what degree and at what speed beta-amyloid (25-35) can aggregate in vitro. It was found that 100 nmol beta-amyloid (25-35) injected icv did impair LTP in Wistar rats when using the weak but not the strong stimulation protocol (P < 0.001). One-hundred nano mole of the reverse sequence amyloid (35-25) had no effect. LTP in Lister Hooded rats was not impaired by amyloid at any stimulation protocol. The aggregation studies showed that amyloid (25-35) aggregated within hours, while amyloid (35-25) did not. These results show that the genetic background and the stimulation protocol are important variables that greatly influence the experimental outcome. The fact that amyloid (25-35) aggregated quickly and showed neurophysiological effects, while amyloid (35-25) did not aggregate and did not show any effects indicates that the state of aggregation plays an important role in the physiological effects.
