[Immunity to embryonal stage-specific antigens in viral carcinogenesis].

The oncogenesis induced by the monkey SA7(C8) adenovirus in CBA/Ca mice has shown that immune responses to embryonal antigens are formed at early stages of the latent period and are preserved for a long time reaching the maximum by the 30th day of the latent period. The observed immune response to early embryonal antigens is considered as a factor of the tumour growth immunostimulation and also as a condition limiting the antitumour immunity.

[Proliferative activity of cultures of embryonic mouse fibroblasts, infected by CA7(C8) adenovirus in a non-serum medium].

Primary monolayer cultures of mouse fetal cells were grown in serum-depleted medium. Cell proliferation parameters were measured by scanning cytophotometry and 3H-thymidine autoradiography. Both 1 h and 24 h thymidine labelling indices (LI) were extremely low in cultures growing without serum for 1 day indicating only residual proliferative activity. The number of cytophotometrically detected S-phase cells was slightly higher than the corresponding LI values. This suggests either the arrest of the cells in S-phase or failure to utilize exogenous thymidine without serum. 24 h after infection by oncogenic adenovirus SA7 (C8) a burst of proliferative activity was revealed in serum-depleted cultures with a dramatic increase of both LI values and the amount of S-cells detected by cytophotometry. These data are discussed in terms of possible virus-induced neoplastic cell transformation.

[Level of cyclic nucleotides (cAMP and cGMP), ATPase activity and calcium ion transport in hamster lymphocytes during tumor growth].

The biochemical systems of lymphocytes of the peripheral blood and lymph nodes of hamsters with a herpes simplex (type 2) virus--induced transplantable tumor Rapp HSV2 were studied. The cAMP concentration in tumor-bearing hamsters was 1.5-2 times higher, while cGMP level--2.2 times lower than in control. This was matched by a rise in the level of transport ATPases in the lymphocytes of experimental animals. Concanavalin A had practically no effect on the uptake of Ca2+ by lymphocytes of tumor-bearing animals. The stimulating effect on the blasttransformation of those lymphocytes was slight. It is suggested that the said biochemical changes in their totality inhibit the activation of lymphocytes in the tumor-bearing organism.

[Cytophotometric study of changes in the structure of nuclear chromatin induced by the oncogenic adenovirus SA7 (C8)].

Infection of primary murine embryonic cell cultures by adenovirus SA7 (C8) results in an increase in chromatin condensation Average optical density of Feulgen stained nuclei 24 h following virus absorption increased for G0/1, S, and G2 cells by 16.1, 11.3 and 13.1%, respectively. This phenomenon is associated with the stimulation of proliferation, with an increase of the S cell amount by 50% of the control values and a decrease of average cell nuclei areas in all phases of cell cycle.

[Changes in the activity of oxidative enzymes in thymus cells of CBA mice during carcinogenesis induced by simian adenovirus SA7(C8)].

Microspectrophotometry was used to study the activity of oxidative enzymes in the cortical and medullar regions of the thymus, in epithelial stromal cells of CBA mice at varying time of postnatal development and during induced virus cancerogenesis. It was shown that in the thymus of control animals, the activity of NAD-, NADP-diaphorases and beta OBDH reached maximum on the 30th day of postnatal development. The animals exposed to virus manifested a distinct lowering of the activity of the enzymes even in the latent period of cancerogenesis. Variations of the activity of the test enzymes during postnatal development of control animals and at varying time of cancerogenesis appear to be related to functional potencies of the thymus.

[Effect of the physiological status of culture on herpes simplex virus type 2 infection of synchronized cells BNK-21].

Infecting of synchronized cells-BNK-21 by herpes simplex virus type 2 in the S period resulted in a secondary rise of the labeled cells index and shortened time of the cell cycle as a whole. While infecting of the cells during the S period some morphological transformation was observed. The infecting of the cells in the periods M, G1, G2 revealed no essential changes in cell DNA synthesis and failed to result in any cell transformation.

[Changes in the cyclic AMP level in the cells of murine sarcoma induced by monkey adenovirus SA7(C8) during tumor growth enhanced by lymphocytes from intact syngeneic mice].

Under consideration is the problem of changes in the cAMP level in sarcoma of mice CBA with routine and enhanced rate of growth. To this end the kinetics of sarcoma growth, induced by monkey adenovirus SA7 (C8), with common and stimulated by lymphocytes from intact syngeneic animals rates of growth was studied. Simultaneously, the kinetics of intracellular cAMP content in tumor cells was studied too. It was found that there is an inverse dependence between the cAMP content and the rate of sarcoma SA7 (C8) growth. Use of one type of tumor cells with the predetermined different rate of growth makes it possible to relate the changes in the content of intracellular tumor cell cAMP with changes being due to growth potentials.

[Proliferation of hamster embryo fibroblast cultures infected with the herpes simplex virus].

In the HEF system in logarythmic and stationary phases of the culture growth the type-2 herpes simplex virus serves as a stimulator of proliferative processes in the cell (there is an increase of the labelled cells index, mitotic index and the intensity of labelling). The capacity of herpes virus not to suppress the cell DNA synthesis, as infestious viruses do, but to stimulate poliferative abilities of the cell indicates a presumable potential oncogenicity of this virus.

[Mechanisms of changes in the mass of the organs central to immunity during adenovirus carcinogenesis].

The spleen cells transfer from mice CBA at the 25th day of the carcinogenesis latent period induced by adenovirus SA7(S8) to newborn syngeneic animals caused the graft versus host reaction in them. There was splenomegaly and progressive decrease in weight of the recipients' thymus. Analogous alterations of lymphoid organs were noted in the animals infected during the neonatal period by oncogenic adenovirus SA7(C8). Results showed that adenoviral carcinogenesis had some manifestations of autoimmune disease.

[Changes in lactate dehydrogenase isoforms in the process of oncogenesis].

Isoenzymes of lactate dehydrogenase were studied by disc-electrophoresis in polyacrylamide gel, and in the clinic--in 1% agar gel. Oncovirus A12 invasion of the culture of rat embryo fibroblasts (REF) was found to result in the increased percentage of the cathode fractions activity (LDG-4 and LD-5) and in the disappearance of LDG-1 yet during the first day of the experiment prior to hypoxia and enhanced proliferation, i. e. it is most likely to be primary. In the homogenates of cancerous tumor and large intestine polyps of man also a reliable increase of the cathode and a decrease or disappearance of the anode fractions accur. A correlation of the experimental and clinical data allowed a suggestion to be made that LDG isoenzymes changes are genetically conditioned and play an important role in the process of oncogenesis, providing conditions for the increased intensity of glycolysis and proliferation.

Isozymic forms of some energy metabolism enzymes during oncovirus-induced cell transformation.

Kinetic studies of changes in isozymic forms of hexokinase, lactate and malate dehydrogenase and in total hexokinase activity during viral carcinogenesis were carried out. The test systems were rat embryo fibroblasts infected with an oncogenic variant of human adenovirus type 12 and an infectious adenovirus type 6, intact REF cultures in different stages of growth (log and stationary phase), and hamster sarcoma A12 and rat reticulosarcoma 321-RRS cell cultures. Molecular isozymic forms of the stated enzymes and total hexokinase activity in the nuclear fraction and cytoplasm of cells in culture were investigated. It was shown that infectious and oncogenic viruses evoked a rearrangement in the spectrum of the energy metabolism enzymes in the nucleus and cytoplasm. The changes appeared in the first days of the contact of REF culture with the virus, and were more pronounced after the oncogenic rather than the infectious virus. The analysis of changes in isozymic forms of the enzymes under study in virus A12-infected REF cultures and in hamster sarcoma A12 and reticulosarcoma 321-RRS cells growing in vitro revealed that they had some features in common. The most pronounced changes were found with hexokinases. The changes described can serve as objective signs of cell transformation.

[Immunostimulation of the growth of a syngeneic sarcoma primarily induced in mice by simian adenovirus SA7(C8)].

Transplantation of splenic cells from CBA mice bearing primary sarcoma SA7(C8) together with autologous sarcoma cells (in the ratio 1 : 1 and 10 : 1) into syngeneic recipients, irradiated with 600 rad, resulted in a marked decrease of the latent period of tumor development. Syngeneic splenocytes of normal mice would enhance the tumor growth with the ratio 10 : 1. Preliminary treatment of tumor-bearing and normal mice with hydrocortisone in the dosage of 6 mg per 100 g of weight fails to reduce the capacity of splenic cells to induce immunostimulation of tumor growth.

[Changes in the isoenzymatic spectrum of energy metabolism enzymes and in the mitochondrial ultrastructure in viral carcinogenesis].

The dynamics of redistribution of the isoenzymatic spectrum of lactate and malate dehydrogenases in the cells of rat embryo fibroblasts (REF) having been in contact with human adenovirus type 12, as compared to the dynamics of changes in the fine structure of the cellular mitochondria of the same models was studied. The action of oncogenic virus caused rearrangment in the isoenzymatic spectra of lactate and malate dehydrogenases both in the nuclei and in the cytoplasm. These disturbances occur from the first days of the REF culture interaction with the oncogenic virus and are not connected with the proliferative growth of the cell culture, but they are stipulated by the very fact of the virus influence on the cell. Comparison of the kinetics of the changes in the enzymes under study with the data of quantitative analysis of the fine structure of the mitochondrial apparatus demonstrated that biochemical shifts occurred much earlier, practically from the moment of infecting.
