Crystallization of human oxyhaemoglobin from poly(ethylene glycol) solutions.

Crystals of human oxyhaemoglobin were obtained from poly(ethylene glycol) solutions. Spectroscopic and spectrophotometric measurements on the solutions during crystallization and on the dissolved crystals indicate that the method yields stable crystals of oxyhaemoglobin. Preliminary X-ray studies showed that the crystals obtained are isomorphous with those of deoxyhaemoglobin obtained from poly(ethylene glycol) solutions [Ward, Wishner, Lattman & Love (1975) J. Mol. Biol. 98, 161-177].

Pergolide mesylate, a dopaminergic receptor agonist, applied with L-DOPA enhances serum antioxidant enzyme activity in Parkinson disease.

The aim of this study was to compare patients with Parkinson disease (PD) patients treated with pergolide mesylate (PM), a dopaminergic receptor agonist, together with L-DOPA and those these treated with L-DOPA alone on the concentration of free radicals (FR), glutathione, and the activity of superoxide dismutase (SOD) and catalase in the serum. The study was carried out using 16 age-matched control subjects, 16 PD patients treated with L-DOPA at a dose of 1 to 1.5 g daily, and 16 PD patients treated with L-DOPA 1 to 1.5 g daily with PM 0.75 to 1.25 mg daily. The mean duration of treatment of PD was 6 years (range, 2-8 years) with l-DOPA, and 2 years with PM + L-DOPA or L-DOPA alone. Although there was no significant difference in lipid peroxidation products among the 3 groups, patients treated with L-DOPA showed high levels of FR as determined by dichlorofluorescein. Although catalase and SOD activities were elevated in both groups of PD patients, additional treatment with PM further enhanced catalase activity compared with those treated with l-DOPA alone. Interestingly, patients treated with PM + L-DOPA showed a significantly increased level of glutathione compared with those treated with L-DOPA alone. Collectively, these data suggest that PM + L-DOPA is a more efficient therapy in maintaining an antioxidative defense in PD patients compared with treatment with L-DOPA alone.

[Assessment of regional blood flow in patients after mild head trauma].

Single photon emission computerized tomography (SPECT) using radioisotope markers was applied for analysis of regional blood flow in the brain. The method makes possible demonstration of the presence and extent of damage to the nervous tissue caused by trauma. The degree of blood flow impairment is of prognostic significance in cases of cerebral concussion. In the present study the regional brain blood flow was assessed shortly after trauma and one year in cases of mild cerebral concussion. In the studied group immediately after trauma blood flow impairment was found mostly in the left temporal area and in frontal areas. Changes of rCBI were present also late after trauma. In 7 out of 16 cases SPECT image failed to change one year after trauma. In 8 cases the rCB improved up to normalization. In 1 case the changes progressed, in another case hyperperfusion focus changed to hypoperfusion. In 2 cases the pattern was normal early and lata after trauma. In the studied group in cases with changed rCBF no changes were found in CT and MRI examinations. This evidence a greater sensitivity and usefulness of SPECT in the assessment of early and late consequences of head trauma.

[The role of nitrogen oxide in migraine attack].

Views are surveyed on the role of nitrogen oxide in the pathogenesis of migraine attacks. There is some evidence that molecule plays an important role in these attacks through vasodilation in the central nervous system. In the process of nitrogen oxide release glutaminergic receptors of NMDA type participate modulating the activity of serotoninergic receptors. Increased synthesis of nitrogen oxide can follow spreading cortical ischaemia, histamine release from mast cells in the meninges and also through activation of the trigeminovascular system. The hypothesis of active role of NO in the pathogenesis of migraine attacks is supported by the analysis of the mechanism of action of drugs effective in the treatment of that disease.
