Core region of Citrobacter O23 lipopolysaccharide. Structure elucidation by chemical methods, gas chromatography/mass spectrometry and NMR spectroscopy at 500 MHz.

A novel enterobacterial core region in Citrobacter O23 lipopolysaccharide is described. Its structure was determined by methylation analysis/mass spectrometry, chemical degradation and one- and two-dimensional 1H-NMR spectroscopy: [formula; see text] where PPEtN stands for diphosphorylethanolamine, and dOclA for 3-deoxy-D-manno-octulosonic acid.

Characterization of an enterobacterial common antigen (ECA) epitope recognized by anti-ECA-tetanus toxoid conjugate serum.

The antigenic reactivity of both native and chemically modified enterobacterial common antigen (ECA) with anti-ECA-tetanus toxoid (TT) conjugate serum was investigated. The results obtained suggest that reduction of the carboxyl group of the mannosaminuronic acid component of ECA diminishes, but does not destroy its antigenic reactivity. Each of the sugar components were found to contribute to its reactivity with anti-ECA-TT conjugate serum, indicating that the trisaccharide repeating unit represents the ECA epitope. A nonasaccharide (trimer of the ECA repeating unit) inhibited antibody binding better than the hexasaccharide dimer, a finding which suggests that oligosaccharide conformation also makes a contribution to its inhibitory activity.

Saccharide-protein covalent conjugates: immunochemical characterization of Citrobacter 036 core oligosaccharide-tetanus toxoid conjugates.

Core oligosaccharides (complete and incomplete) isolated from Citrobacter 036 lipopolysaccharide were covalently conjugated with tetanus toxoid. Serological examination of the Citrobacter 036 core oligosaccharide-tetanus toxoid conjugates showed that they are strong immunogens. The monospecific anti-conjugate sera prepared by immunization of rabbits, were used to study the antigenic relations between lipopolysaccharide core regions of 8 strains of Citrobacter. Immunoelectrophoresis, immunoblotting and quantitative microprecipitation were performed in the experiments.

Detection of common polysaccharide antigen of Pseudomonas aeruginosa with O-antiserum to Pseudomonas cerasi.

The identity of the structures of common polysaccharide antigen (CPA) of Pseudomonas aeruginosa and O-antigen of Pseudomonas cerasi was used for immunochemical study of polysaccharide antigens of seven immunotypes (IT) of P. aeruginosa. ELISA performed with O-antiserum to P. cerasi showed that CPA is present in all seven ITs in different amounts. In SDS-PAGE this antigen behaves as a lipopolysaccharide (LPS) and is detected by immunoblotting technique in five of seven ITs.

[Activity of natural killer cells (NK) in patients with atopic diseases of the respiratory system].

NK cells are one of the crucial elements of natural cellular immunity mechanisms in situations when specific mechanisms are not altered. The aim of this study was to study activity of such cells in atopic respiratory diseases. The study was carried out in 52 atopic patients; 37 had atopic respiratory diseases, 15 atopic dermatitis. Control consisted of 28 healthy subjects without a history of atopy. Effector lymphocytes were recovered after centrifugation on the Ficoll/Uropolin gradient from venous blood. The activity of NK cells was determined using the cytotoxic test studying the level of emitted 51Cr from a standard K562 cell line. The authors have shown that the activity of NK cells is increased but this does not correlate with the level of IgE. This was seen mostly in patients with atopic respiratory diseases. In subjects with atopic dermatitis the activity of NK cells was similar to the seen in control subjects.

The structure of the biological repeating unit of the O-antigen of Hafnia alvei O39.

On mild acid-catalysed degradation of the lipopolysaccharide from Hafnia alvei O39 followed by gel filtration of Sephadex G-50, the O-specific polysaccharide and three oligosaccharides were obtained, which represent the core substituted with 0-2 O-antigen repeating-units. On the basis of sugar and methylation analyses, 13C-n.m.r. data, solvolysis of the polysaccharide with anhydrous hydrogen fluoride, and computer-assisted 13C-n.m.r. analysis of the Smith-degraded polysaccharide, it was concluded that the biological repeating unit of the O39 antigen was Formula; see text

O-specific polysaccharides of Hafnia alvei lipopolysaccharides isolated from two serologically related strains: ATCC 13337 and 1187. A serological and structural study using chemical methods, gas chromatography/mass spectrometry and NMR spectroscopy at 500 MHz.

The O-specific polysaccharides of Hafnia alvei ATCC 13337 standard strain and 1187 strain have been isolated and characterized. By means of 1H-NMR spectroscopy, methylation analysis and periodate oxidation, the repeating unit of the polysaccharides could be allocated the respective structures. (formula; see text) where Acyl = D-3-hydroxybutyryl, and 3-O-acetylation was to about 66%. The structural similarity of the polysaccharides was confirmed in the serological study; their epitopes were determined and the importance of various structural elements for the serological specificity was discussed.

Structure elucidation of the core regions from Citrobacter O4 and O36 lipopolysaccharides by chemical and enzymatic methods, gas chromatography/mass spectrometry, and NMR spectroscopy at 500 MHz.

Novel enterobacterial core oligosaccharides were isolated from Citrobacter O4 and O36 lipopolysaccharides, and their structures were determined by methylation analysis, Smith degradation and enzymatic degradations, gas chromatography/mass spectrometry, and two-dimensional phase-sensitive correlated, relayed coherence transfer, double-quantum, triple-quantum-filtered, and nuclear Overhauser effect (NOE) 1H NMR spectroscopy at 500 MHz. In the formulas, all hexose residues are D-hexopyranoses, and heptoses are L-glycero-D-manno-heptopyranoses; the alternative locations of the side-chain heptose and pyrophosphorylethanolamine (PPEtN) residues are marked by dashed lines; dOclA stands for 3-deoxy-D-manno-octulosonic acid. (formula; see text) Along with these complete cores, incomplete ones, lacking the hexosamine trisaccharides, occur in the lipopolysaccharides of both types. Qualitative NOE data were in good agreement with the minimum energy conformation of the above O36 oligosaccharide, calculated with the aid of the SUGAR program [Sundin, A., Carter, R. E., & Liljefors, T. (1988) J. Mol. Graphics (in press)].

Hafnia alvei lipopolysaccharides: isolation, sugar composition and SDS-PAGE analysis.

Lipopolysaccharides (LPS) of 33 strains of Hafnia alvei were isolated and purified. LPS content of the dry bacterial mass ranged from 1.2 to 4.5%. All examined lipopolysaccharides contained glucose, glucosamine, heptose, 3-deoxy-octulosonic acid and often galactose. Rhamnose, mannose, galactosamine, mannosamine and unidentified amino sugars were found in some H. alvei strains. Sialic acid was present in LPS of one strain. D-3-Hydroxybutyryl groups also were identified in lipopolysaccharides of 5 strains of this genus. SDS-PAGE of the lipopolysaccharides was presented in the paper. According to these results two core types exist in H. alvei.

Structure determination of the O-specific polysaccharides from Citrobacter O4- and O27-lipopolysaccharides by methylation analysis and one- and two-dimensional 1H-NMR spectroscopy.

Using sugar and methylation analyses, and one- and two-dimensional 1H-NMR spectroscopy at 500 MHz it was established that poly-beta-1,2-4-deoxy-D-arabinohexopyranose occurs as O-specific chains of lipopolysaccharides in Citrobacter serotypes O4, O27 and O36. Strong serological cross-reactivity between these serotypes is in full agreement with the chemical identity of their O-specific polysaccharides.

Characterization and diagnostic application of a lipopolysaccharide core oligosaccharide-protein conjugate.

Shigella sonnei phase II core oligosaccharide (OSPhII) was linked covalently with protein (bovine serum albumin or tetanus toxoid) to obtain a conjugate (OSPhII-protein) of good immunogenicity in rabbits. Anti-OSPhII-protein conjugate sera contained high levels of immunoglobulin G antibodies against the complete core region of Sh. sonnei lipopolysaccharide. The antigenic relationships between lipopolysaccharides of R1, R3 and R4 core types using anti-OSPhII-protein sera were studied. These antisera may be used for the rapid determination of core types in unknown enterobacterial strains.

Core region of Citrobacter lipopolysaccharide from strain PCM 1487. Structure elucidation by two-dimensional 1H-NMR spectroscopy at 500 MHz and methylation analysis/mass spectrometry.

The core structure of Citrobacter PCM 1487 lipopolysaccharide has been established using methylation analysis/mass spectrometry, chemical degradations and one- and two-dimensional 1H-NMR spectroscopy at 500 MHz. 1H-NMR assignments are given for all sugar components of the core oligosaccharide. In the formula shown below, the alternative locations of branch terminal heptose (LDHep) and diphosphorylethanolamine (PPEtN) residues are marked by dashed lines; dOclA stands for 3-deoxy-D-manno-octulosonic acid. (Formula: see text). The sample of the core oligosaccharide showed some microheterogeneity due to a slightly incomplete substitution by terminal N-acetylgalactosamine and a partial splitting of diphosphorylethanolamine residues.
