E. coli recA protein possesses a strand separating activity on short duplex DNAs.

RecA protein was found to catalyze the dissociation of the strands of a DNA substrate consisting of a 20-nucleotide primer annealed to circular single-stranded M13mp DNA. The strand separation reaction requires ATP hydrolysis and the presence of single-stranded DNA flanking the duplex DNA region to be unwound. RecA-catalyzed strand separation is effective only for very short duplexes, not exceeding 30 bp, and is not stimulated by single-stranded DNA-binding protein. These results are consistent with the ability of recA protein to disrupt regions of secondary structure in single-stranded DNA and to incorporate large non-homologies into heteroduplex DNA.

Pain as a symptom in elderly depressed patients. Relationship to diagnostic subgroups.

The presence of pain as a symptom has been studied in a series of 51 depressed elderly inpatients and in a control group of 71 subjects. The frequency of patients with moderate to severe pain was significantly higher in the experimental group (72%) than in the controls (33.8%). Of the various categories diagnosed according to the DSM III degree criteria, the highest scores for pain were gained by the subjects suffering from dysthymic disorder and atypical depression, while those obtained by the patients with major depression and adjustment disorder with depressive mood were lower. The difference does not seem to depend on the quantity of anxiety present.

Ranitidine versus ranitidine and prazepam in the short-term treatment of duodenal ulcer--a double-blind controlled trial.

Fifty out-patients with an active, endoscopically proven duodenal ulcer were entered a double-blind trial of ranitidine + prazepam or ranitidine + placebo. Two drop-outs occurred in the prazepam group and 1 in the placebo group. After 28 days of treatment, the ulcer had healed in 95.6% of the patients on ranitidine + prazepam and 75% of the patients on ranitidine + placebo (p = 0.03). Sleepiness was the most frequent side effect, reported by 8 subjects in the prazepam group and by 1 subject in the placebo group. It is concluded that prazepam can be usefully combined with ranitidine in the short-term treatment of duodenal ulcer.

Evaluation of use patterns of psychotropic drugs in an Italian geriatric hospital.

This study is a survey of psychotropic drug use patterns among 934 elderly patients admitted to the Padua Geriatric Hospital. 34% of the patients took psychotropic drugs during their stay in hospital. The drugs most commonly used are anxiolytics and hypnotics of the benzodiazepine class. Women subjects living alone, and to a lesser extent those suffering from particular disorders, as of the central nervous system and the musculoskeletal apparatus, seem to be at higher risk in the use of psychotropic drugs. Moreover, the results show that psychotropic drugs are prescribed during the stay in hospital and even on discharge to a high percentage of subjects who did not use such drugs before admission to the hospital.

A case of Capgras delusion presenting as a postpartum psychosis.

A case is reported of Capgras syndrome arising as the major symptomatologic manifestation of a postpartum psychosis. Other diagnostic investigations have called into question the most recent etiopathogenetic interpretations which preclude the concomitant existence of an organic disorder, previously held to be a precondition for the development of Capgras syndrome.

Gastric emptying and anxiety in chronic duodenal ulcer.

The authors have evaluated in a group of patients afflicted with duodenal ulcer whether anxiety might be in some way linked to gastric emptying. The 23 patients studied were subdivided into two groups: "fast" emptiers (n = 12; t 1/2 less than 90 min) and "normal" emptiers (n = 11; t 1/2 greater than 90 min). There were no significant differences between the two groups for the following parameters: sex, age, marital status, educational level, social class, number of cigarettes smoked per day, type of treatment undergone for the duodenal ulcer and maximal acid output. Anxiety levels measured with the Q4 and QII factors of the Cattell's PF 16 were found to be the same in the two groups. The authors conclude that anxiety does not seem to be linked to gastric emptying in chronic duodenal ulcer.

Psychotropic drug use in an Italian general hospital.

The present study is a survey of psychotropic drug use patterns among 1860 patients admitted to Padova General Hospital in North-East Italy. Our results show that 23% of the patients took psychotropic drugs for almost their entire stay in hospital. The drugs most commonly used are anxiolytics and hypnotics, both of the benzodiazepine class. The use of barbiturates as hypnotics has almost disappeared. Some significant correlations between the use of psychotropic drugs and socio-demographic factors were also found. Women, widows and widowers, retired and elderly persons seem to be at higher risk in the use of psychotropic drugs. The meaning and implications of these results are discussed.

Chronic low-back pain and depression: an epidemiological survey.

The author evaluated the presence of depression among 64 male workers at an engineering factory, suffering from chronic low-back pain (19 organic cases and 45 idiopathic cases). Moreover, the frequency of depressive spectrum disorders was assessed among their first degree relatives. No significant differences emerged between organic and idiopathic cases.

Chronic pelvic pain and depression.

The psychological and clinical characteristics of 29 women who underwent laparoscopy for chronic pelvic pain (CPP) have been studied. Patients (n = 11) with CPP without obvious organic pathology showed a significantly higher mean score at the Zung Self-Rating Depression Scale and more somatic symptoms which usually accompany depression than patients (n = 18) with CPP in whom pathology was found; they also showed higher frequencies for depressive disorders in personal anamnesis and for 'depression spectrum disorders' in family history. No differences between the two groups emerged for anxiety, somatization and inadequacy as measured by the Kellner-Sheffield Symptom Rating Test. Through a comparison with a group of women affected with another type of idiopathic chronic pain, and on the basis of the above reported results, the authors suggest that CPP without obvious organic pathology might be an expression of a depressive disorder.

Personality and psychological factors in chronic duodenal ulcer. Their interaction with biological parameters.

Forty patients with chronic duodenal ulcer were studied to determine whether some particular personality and psychological factors are associated with this disease. To this purpose the Cattell's PF 16 personality Test was utilized. Duodenal ulcer population presented a typical personality profile characterized by: high anxiety, dependence and introversion. Some biological parameters, suggested as predictors of slow healing and/or relapse of ulcer, were studied: patients with maximal acid output greater than 60 mEq/h and /or total serum pepsinogen greater than 109 micrograms tyr/ml/24 hrs and/or serum pepsinogen group I greater than 99 ng/ml showed an anxiety level significantly higher than that of patients with values less than the reported thresholds. These results suggest a contributory role by psychological factors in duodenal ulcer disease. The association of anxiolitic drugs to the traditional antiulcer therapy may be proposed, particularly in hypersecretors.

Uridine phosphorylase from Escherichia coli B.: kinetic studies on the mechanism of catalysis.

Using a highly purified enzyme preparation of uridine phosphorylase from Escherichia coli B, we have performed detailed kinetic studies which include initial-velocity and product-inhibition experiments in the forward and reverse directions of the reaction. These studies indicate a rapid-equilibrium random mechanism for this enzyme with the formation of an enzyme . uracil phosphate abortive complex. Lack of formation of the enzyme . uridine . ribose-1-phosphate abortive complex suggests that the ribosyl moiety of the two ligands compete for the same binding site. The random mechanism is different from the ordered addition of substrates found for uridine phosphorylase from other sources. All the kinetic constants in the forward and reverse directions and the Keq of reaction for E. coli uridine phosphorylase are reported herein.
