[Primary MALT-type malignant lymphomas of the salivary glands].

Six cases of primary extranodal lymphomas in big salivary glands, which met criteria of MALT lymphoma, were investigated with the aid of antibodies against Ig, light chains, LCA and EMA, in addition in two cases against Ig heavy chains and with antibodies KL-1 and VCHL-1. All the tumours had centrocytoid morphological features, one of them showed signs of focal blastic transformation into centrocytoid centroblastoma. Tumour cells showed twice plasmacytic and three times plasmacytoid differentiation with intracytoplasmic Ig monoclonality (once IgM/kappa, once IgA/kappa, three times kappa positivity). They were positive in reaction against epimyoepithelial proliferation. A picture of myoepithelial sialoadenitis with partial lymphomatous infiltration prevailed in two cases and a lymphoma picture with remnants of sialoadenitis in three cases. Features of inflammatory process were lacking in a case with blastic transformation. There is a dispute whether features of sialoadenitis belong to the defining criteria of MALT type lymphoma or can disappear during blastic transformation of a low grade malignant lymphoma.

[Prognostic relevance of histopathologic subtyping of mixed-cell Hodgkin's disease].

Mixed cellular (MC) type of Hodgkin disease was studied in a group of 92 bioptic cases diagnosed before the start of treatment. A precise histological analysis showed one subtype with lower malignancy (MC 1) and a second one with higher malignancy (MC 2). Subtype MC 1 comprised 61 cases (66 per cent), subtype MC 2 31 cases (34 per cent). Histomorphological subtyping was brought into correlation with clinical data of survival available in 65 cases (44 cases of subtype MC 1,21 cases of subtype MC 2). Different survival in both subtypes had statistical significance and prognostic relevance. Suggested subtyping is simple, based on common histological slides, without need of special additional investigation.

[Crohn's disease of the stomach].

The authors describe the incidence of Crohn's disease at an uncommon site the stomach--in a 40-year-old man. With regard to the steadily increasing incidence of the disease, it is important to consider its presence also in uncommon sites in the gastrointestinal tract. The establishment of the diagnosis must be comprehensive, as even histological examination of endobioptic specimens or resected portions need not be unequivocal from the diagnostic aspect.

[Prognostic relevance of histopathologic subtyping of nodular-sclerotic types of Hodgkin's disease].

The paper analyzes the histopathologic pattern of 30 bioptically assessed cases of nodular-sclerotic (NS) type of Hodgkin's disease diagnosed before onset of therapy. The ratio of lymphocytes and tumor cells, as well signs of atypia, pleomorphism and blastic proliferation of tumor cells were evaluated in each case. On the basis of this analysis, the NS type was divided into the subtype NS 1 (14 cases) and subtype NS 2 (16 cases). In the subtype NS 1 lymphocytes were predominant in the tumor nodes and there were only few Sternberg-Reed cells, which were either isolated or in small clusters. Fibrosis in the tumor nodes was mild. In the subtype NS 2 the ratio of lymphocytes was lower, Sternberg-Reed cells were numerous and were found to be in groups and clusters. Pleomorphism and atypia of the tumor cells was remarkable. Pronounced fibrosis was seen in the nodes. The ratio of reactive cells and necroses exhibited individual variation in both subtypes. Statistical evaluation of survival time showed significant differences between the patients with subtype NS 1 and NS 2. Differentiation between NS 1 and NS 2 subtypes of the nodular-sclerotic type of Hodgkin's disease can thus ben considered to be prognostically relevant.

Effects of atropine, acetylsalicylic acid, FPL 55712, and phentolamine on increased histamine responsiveness of cat lung strips under conditions of airway inflammation.

Our recent in vitro studies on airway smooth muscles of cats with turpentine oil inflammation showed an increase of isometric tension of the lung strips to histamine application. This communication describes the effect of atropine, acetylsalicylic acid, FPL 55712, and phentolamine on the histamine contractions of the lung strips derived from control and experimental groups of cats. Pretreatment of the lung strips with atropine and acetylsalicylic acid had no significant effect on histamine induced contraction. FPL 55712 significantly decreased the mean values of isometric contractions after the low doses of histamine in experimental groups of strips. The isometric contractions after higher doses of histamine were not affected by FPL 55712 in both groups of strips. The significant increase of histamine contractions of the lung strips induced by experimental inflammation was reduced by phentolamine. The role of alpha adrenergic receptors in the increased responsiveness of the inflamed lung tissues to histamine is discussed.

Acanthamoeba profilin binding to fluorescein-labeled actins.

The binding constants of Acanthamoeba profilin to fluorescein-labeled actin from Acanthamoeba and from rabbit skeletal muscle have been determined by measuring the reduction in the actin tracer diffusion coefficients, determined by fluorescence photobleaching recovery, as a function of added profilin concentration. Data were analyzed using a two-parameter nonlinear regression analysis to determine the profilin-actin dissociation constant Kd and the profilactin diffusion coefficient, DPA. For fluorescein-labeled Acanthamoeba actin, the least-squares estimates for Kd and DPA, along with approximate single standard deviation confidence intervals, are Kd = 48 (36, 63) microM and DPA = 6.72 (6.62, 6.81) X 10(-7) cm2s-1. For fluorescein-labeled skeletal muscle actin, the corresponding values are Kd = 147 (94, 225) microM and DPA = 6.7 (6.3, 7.0) X 10(-7) cm2s-1. These dissociation constants are the first to be determined from direct physical measurement; they are in agreement with values inferred from earlier studies on the effect of profilin on the assembly of actin that had been fluorescently labeled or otherwise modified at Cys 374. These results place important restrictions on the interpretation of experiments in which fluorescently labeled actin is used as a probe of living cytoplasm or cytoplasmic extracts that include profilin.

Assembly of Acanthamoeba actin in the presence of Acanthamoeba profilin measured by fluorescence photobleaching recovery.

Assembly of Acanthamoeba actin, of which trace quantities had been labeled with 5-(iodoacetamido)-fluorescein, was quantified using the modulation detection method of fluorescence photobleaching recovery (FPR). This technique permits explicit determination of the fraction of labeled actin incorporated into filaments and the translational diffusion coefficients of the filaments, from which filament length can be calculated. Addition of Acanthamoeba profilin in molar ratios to actin of about 1.1:1 and 2.3:1 retarded the initial kinetics of assembly (induced by addition of 2mM Mg+2) and reduced the fraction of actin incorporated into filaments. The diffusion coefficients of filaments formed were greatly changed by the presence of profilin at short times, but the differences became increasingly smaller at longer times. After 26 hr. the filaments formed in 1.1:1 profilin were about 12% shorter and in 2.3:1 profilin were about 20% shorter than filaments formed by actin alone under the same conditions.

Study of cough in anaesthetized cats with experimental pulmonary oedema.

The authors studied parameters of laryngopharyngeal (LPh) and tracheobronchial (TB) cough (the number of efforts--NE, the number of efforts per minute--NE.min-1, the intensity of the maximum effort--IME and the intensity of the attack--IA), respiratory parameters (VT, f, Cdyn), blood gas values (PaO2, PaCO2) and the pH in the arterial blood of anaesthetized cats (Pentobarbital Spofa, 35 mg x kg-1 i.p. with experimental pulmonary oedema. Oedema was induced by the i.v. administration of a fatty acid mixture (capric acid 3.8 g, caprylic acid 3.1 g and olive oil 3.0 g)--in a dose of 0.05 ml x kg-1 in group 1 and of 0.01 ml x kg-1 in group 2. Cough was elicited by stimulating the laryngopharyngeal and tracheobronchial mucosa with a silon fibre. Pulmonary oedema was determined from a macroscopic and microscopic examination of the lungs, from the ratio of lung weight to body weight and from the percentual dry weight of the right lungs. The authors found that the intensity of cough was depressed during pulmonary oedema, particularly in the case of TB cough. VT and Cdyn fell, f rose and the PaO2 and pH values steadily decreased.

Histopathologic analysis of the so-called secondary blastic non-Hodgkin's malignant lymphomas.

Fifteen cases of blastic transformation of originally low grade malignant lymphomas to high grade malignant ones were studied. The material was selected from the biopsy registry of our institute and had been collected over the period of 1 January 1982 to 1 March 1984. In 10 cases this transformation was verified morphologically (in 8 cases by successive biopsies, in 2 cases in the sectional material). At the time of the first biopsy, transformation was suspected in four cases. There were 4 cases of transformation of centroblastic-centrocytic lymphomas to centroblastomas, one case of centrocytoma transformed to centroblastoma, two cases of immunocytomas transformed to centroblastomas, and three cases of immunocytomas transformed to immunoblastomas. The transformation took place within a mean time of 9.5 months after the first biopsy, and 6 patients died, on the average, within 5.3 months after the rebiopsy. In live cases only 1 bioptic examination was done on the basis of which the blastic transformation has been suspected, though not proved yet. There was one case of a suspected transformation of a centroblastic-centrocytic lymphoma to a centroblastoma type with structural remnants of low grade malignant lymphomas, in one case the In the further two cases there were blastic malignant lymphomas of the centroblastoma type with structural remnants of low grade malignant hymphomas, in one case the centroblastic-centrocytic type, in the other the immunocytic lymphoma type. The morphological criterion for a suspected rise of a secondary blastic lymphoma is the presence of clear regions composed of an accumulation of blasts with increased mitotic activity in the prevalent structure of the low grade malignant lymphoma.

Effect of sterol incorporation on head group separation in liposomes.

Electrophoretic mobilities of multilamellar liposomes of varying composition have been measured to determine the effect of incorporated sterols on surface charge density. Liposomes made from mixtures of zwitterionic egg phosphatidylcholine (PC) and anionic egg phosphatidylglycerol (PG) in varying proportions were shown to have electrophoretic mobilities consistent with the anticipated surface charge density. Incorporation of cholesterol up to 50 mole per cent in the bilayer produced no detectable change in surface charge density. Similar results were obtained for lanosterol and epicoprostanol. These results are interpreted to mean that incorporation of the sterols into the bilayers produced no detectable change (less than 3%) in the spacing of charged phospholipids. It is inferred that sterols are incorporated among the fatty acyl chains of these phospholipid bilayers with little or no displacement of the head groups at the surface.

[Diagnosis and classification of pulmonary findings in neonates].

Developmental phases could be defined in neonatal lung tissue fixed in situ or in an ordinary way: glandular lung, canalicular lung with simple tubes covered by cuboid epithelial cells, alveolar lung with usual complicated structure. Canalicular lung could manage the blood saturation which depends on the number of capillaries. There was a different picture of alveoli filled with fluid or air. Hyaline membranes, always occurring in fluid-filled alveoli, represented a barrier for drainage of intraalveolar fluid. Step-wise inflation of lung enabled a histomechanical examination giving lately "functional" figures. They showed a poor functional readiness as well as a quick postnatal adaptability of immature lung. Pressure-volume curves correlated with histological maturity and pathological finding offered a more comprehensive explanation of clinicopathological mechanisms.
