Pharmacological evidence for a stimulation of dopamine neurons by noradrenaline neurons in the brain.

The effects of yohimbine (3 mg/kg i.p.), phenoxybenzamine (20 mg/kg i.p.) and clonidine (0.1 mg/kg i.p.) on the synthesis and the utilization of dopamine and noradrenaline in the central nervous system of rats were investigated. Dopa accumulation following decarboxylase inhibition and the alpha-methyltyrosine-induced disappearance of the amines were used as the measure of these effects. The synthesis and the utilization of dopamine and noradrenaline were accelerated by yohimbine. Clonidine plus phenoxybenzamine inhibited the synthesis and utilization of dopamine and the combination also partly antagonized the effects of yohimbine on the trunover of dopamine. The effects of the three drugs on the synthesis and utilization of dopamine might be secondary to their actions on alpha-adrenoreceptors of noradrenaline synapses since, at the doses used, yohimbine increases the release of noradrenaline, phenoxybenzamine blocks postsynapptic noradrenaline receptors and clonidine reduces the release of noradrenaline. This hypothesis is supported by the findings that yohimbine and phenoxybenzamine did not change the increased synthesis of dopamine in reserpine-treated rats and that clonidine did not inhibit the increased synthesis of dopamine after axotomy or treatment with reserpine.

Butyrophenones and brain serotonin metaboism.

The influence of butyrophenone derivatives on the level of serotonin and 5-hydroxyindoleacetic acid in various regions of brain of normal and pargyline-treated rats was investigated. The result indicate that all butyrophenone derivatives used decrease serotonin utilization in rat brain stem while they have smaller influence on this process in the other rat brain regions examined.

Influence of NBTI on metabolism of brain-HT.

The influence of NBTI, an agent which stimulated dopamine receptor in the central nervous system, on concentrations of serotonin and its metabolic, 5-hydroxyindoleacetic acid in the brain of rats was studied. Turnover rate of serotonin in the brain after administration of NBTI to normal rats and those pretreated with spiroperidol, a factor which blocks central dopamine receptors, was evaluated. The changes in metabolism of brain serotonin under the influence of NBTI are similar to those produced by another factor which stimulates central dopamine receptors, namely apomorphine. NBTI causes transient increase in exploratory activity of rats and lowers body temperature.

Influence of dopamine-like compounds on stereotypy and locomotor activity in atropinized rats.

The influence of atropine on stimulation of exploratory activity, locomotor activity and stereotypy in rats treated with compounds which stimulate central dopamine receptors (apomorphine, D-amphetamine, D-145 and piribedil) was studied. Atropine potentiated the action of these compounds in some experimental systems. The results are discussed from the point of view of an equilibrium between the dopaminergic and cholinergic systems in the central nervous system.

Influence of benzodiazepines on turnover of serotonin in cerebral structures in normal and aggressive rats.

The influence of chlorodiazepoxide (CDP), oxazepam (OX), temazepam (TE) and nitrazepam (NI) on serotonin (5-HT) turnover in the brain stem and hypothalamus of normal and aggressive rats was studied on the basis of the rate of disappearance of 5-hydroxyindoleacetic acid (5-HIAA) after inhibition of MAO. Agressiveness in rats was induced by electric stimulation. All the tested compounds diminished 5-HT turnover in both cerebral structures in normal and aggressive rats. The strongest inhibition was produced by CDP in the brain stem of normal and hypothalamus of aggressive rats. The remaining substances acted with equal strength. Only OX had no influence on 5-HT turnover in the brain stem of aggressive rats.

Pharmacologic effects of apomorphine in rats with altered level of brain serotonin.

The influence of inhibitors and precursors of serotonin synthesis on motor stimulation, hypothermia and apomorphine-induced stereotypy was studied. The results are discussed with reference to the previously expressed suggestion of a modulating role of serotonin on the mechanism of apomorphine-induced motility and in apomorphine-induced hypothermia.
