Effect of cadmium upon the activity of some selected marker enzymes in rat liver.

The aim of this study was to determine activities of selected enzymes, i.e. Mg(++)-stimulated ATP-ase, acid phosphatase (AcP), esterase and succinate dehydrogenase (SDH) in rat liver after intragastric administration of cadmium chloride. The investigations were performed on forty male rats, Wistar strain, which made up one control group and two experimental groups. The rats of one experimental group were given a single dose of cadmium chloride solution of 88 mg CdCl2 per kg body weight (Group I), whereas the other experimental group received a double dose, i.e. 176 mg CdCl2 per kg body weight (Group II). The activities of enzymatic reactions were determined by means of analysis of surface optical density using the image computer analyzer, whereas the degree of mental accumulation was estimated with atom absorption spectrophotometry. Following cadmium chloride administration, the activities of SDH and esterase decreased in both experimental groups compared to controls. The decrease in both enzymatic activities was statistically significant. Similarly, activities of ACP and Mg(++)-stimulated ATP-ase decreased below control level in the rats of the Group I, which were given lower dose of CdCl2. This change was not statistically significant. Higher dose of CdCl2 (Group II) caused a statistically significant increase in acid phosphatase activity and only a small increase (bearing no statistical significance) in Mg(++)-stimulated ATP-ase activity. In addition, it was found that the accumulation of cadmium in both the liver and the kidneys is dose-related.

Changes of enzyme activity in the rat liver at different age in circadian cycle.

Most of the biological processes in the living organisms of both animals and man are known to be of rhythmical nature. Variability of enzymatic activity in circadian cycle depends on many factors among other on age, sexual maturity, diet as well as medication. The results obtained in our studies indicate, that the activity changes of acid phosphatase and ATP-ase Mg++ dependent in the liver of all the examined age groups were of 24 hour circadian rythm. As to the acid phosphatase activity the results of this experiments showed that in circadian cycle in all examined age groups there is only one peak of elevated activity. ATP-ase Mg++ dependent showed two activity peaks appearing at the same hour both in 30 and 60 days old animals. It should be noticed that the activities of ATP-ase Mg++ dependent in 100 day old animals were two times higher than in 30 and 60 days old rats.

[Effect of age on the activity of hepatic mixed function oxidases in female rats].

Female Wistar rats aged 5, 10, 15, 30, 60 and 540 days were tested. The rough and smooth microsomes fraction of the liver were tested for protein, cytochromes P-450 and b5 contents as well as for NADPH-cytochrome c reductase, NADH-cytochrome b5 reductase and two monooxygenases activity. Cytochrome P-450 and b5 contents and NADH-cytochrome b5 reductase activity were increasing up to the age of 60 but aniline hydroxylase--up to the age of 15 days. NADPH-cytochrome c reductase activity exhibited a gradual decreasing trend with age. In senescent animals (540 days) the contents of cytochromes and monooxygenases activity were lower but NADPH-cytochrome c reductase activity was higher, as compared to young rats.

[Effect of age on the activity of hepatic mixed function oxidases in female rats after phenobarbital induction].

Studies were performed on young (5-, 10-, 15-, 30- and 60 days old) and senescent (540-days old) female Wistar rats. The rats were administered (i.p.) phenobarbital (150 mg per 1 kg body weight) twice: 72 and 48 h before being killed. In smooth (SER) and rough (RER) hepatic microsomes the activity of mixed functions oxidases was studied. In particular age groups phenobarbital has a changeable effect on the mixed functions oxidases. All the examined components except for the b5 cytochrome content and the NADH-cytochrome b5 reductase activity are induced. In the group of young rats the highest induction was observed in 15- and 30 days old animals and the lowest--in 5- and 60 days old ones. Only for cytochrome P-450 content and NADPH-cytochrome c activity in rough microsomes and protein content in smooth microsomes was the maximal induction highest in 60 days old rats. Small decrease in the protein content in smooth microsomes in 540 days old animals make one suggest the senescent rats are less susceptible to phenobarbital.

Evolution of localization of reactions of adenosine triphosphatase (Mg++-ATP-ase), 5'nucleotidase (5'nt), alkaline phosphatase (AP), and acid phosphatase (AcP) in developing rat testis. II. After CdCl2 treatment.

The described experiments show the influence of a single dose of cadmium chloride (1.5 mg CdCl2/kg body weight applied intraperitoneally) on development of the male gonads from the 1st d of post-fetal life to 1.5 a of life. In the case of the enzymes triggering transportation processes, adenosine triphosphatase stimulated by Mg++ (Mg++-ATP-ase), alkaline phosphatase (AP), and 5'nucleotidase (5'Nt), a considerable damages begin to appear in the 15th d of life whereas in the case of acid phosphatase (AcP) already in the 1st d of life whereas in the case of acid phosphatase (AcP) already in the 1st d of life. These damages increase with age reaching their maximum in the 45th d of life.

Studies on the optimization of parameters of preparative liquid chromatographic columns for production of cardiac glycosides.

Investigations aimed at maximization of the output of the production of cardiac glycosides from their mixtures using liquid chromatography are reported. The dependences of the yield of the process on column length, particle size and surface area of the packing and linear velocity of the mobile phase were examined. It has been established that a simultaneous decrease in the particle size and an increase in the column length and mobile phase flow-rate results in an improvement in the yield. However, such a procedure is limited by technological considerations. Optimum production conditions have been characterized.

Defective natural-killer- and killer-cell activity associated with increased polymorphonuclear leukocyte adherence in psoriasis.

Patients (n = 45) with psoriasis vulgaris were studied for natural killer (NK) cell activity and antibody-dependent cell cytotoxicity (ADCC) of their peripheral blood mononuclear cells against K-562 and L1210 target cells, respectively. Simultaneously, the studies were performed on the adherence to human endothelium of polymorphonuclear leukocytes (PMN) from various forms of the disease. Mean values of NK-cell activity in psoriasis vulgaris, as a whole, and mean values of ADCC in male patients were significantly lower than in a group of healthy controls. This decrease was most pronounced in patients with the active form of the disease, in whom in vitro PMN adherence to human endothelium was found to be increased. Neutrophils from both patients with active psoriasis and healthy individuals were capable of reducing NK-cell activity of normal human mononuclear cells.

Evaluation of localization and intensity of the reactions of succinic dehydrogenase, NADH tetrazole reductase and lactate dehydrogenase in developing rat testis. I. Physiological conditions.

The localization, intensity, and character of the enzymatic reactions of the following enzymes responsible for the oxygenic and nonoxygenic respiration were under examination: succinic dehydrogenase (SDH), NADH tetrazole reductase (NADH-R), and lactate dehydrogenase (LDH). The features of the enzymatic reactions enumerated above were tested in male gonads of the rats on their 1st, 4th, 7th, 15th, 30th, 45th, 60th, 90th d and 1.5 year of post-fetal life. It was estimated on the examined reactions that their metabolic maturity in the gonad was reached on the 45th d of the rat's life.

Evaluation of localization and intensity of the reactions of succinic dehydrogenase, NADH tetrazole reductase, and lactate dehydrogenase in developing rat testis. II: After cadmium chloride treatment.

The described experiments showed the influence of a single dose of cadmium chloride (1.5 mg CdCl2/kg body mass) applied intraperitoneally on the activity of succinic dehydrogenase (SDH), NADH tetrazole reductase (NADH-R), and lactate dehydrogenase (LDH) in the rat's male gonad. It was ascertained that disturbances of the reaction begin to occur in the 7th d of post-fetal life: they increase gradually, first at the circumference and then the centre of the organ reaching their maximum in 1.5 year old rats.

Evolution of localization of the reactions of adenosine triphosphatase (Mg++-ATP-ase), 5'nucleotidase (5'nt), alkaline phosphatase (AP), and acid phosphatase (AcP) in developing rat testis. I. Physiological conditions.

The experiments were performed upon the rats aged 1, 4, 7, 15, 30, 45, 60, 90 d, and 1,5 a. The behavior of the following reactions was described: for adenosine triphosphatase stimulated by Mg++(Mg++-ATP-ase), for 5'nucleotidase (5'Nt), for alkaline phosphatase (AP), for acid phosphatase (AcP). The first 3 are markers of the transport enzymes in cells, and the 4th is a marker of lytic processes. It was estimated on the basis of the examined reactions that a full metabolic maturity of the gonad was revealed since the 45th d of post-fetal life.

Morphometric studies of development of the rat testis under physiological conditions and after CdCl2 poisoning.

The morphometrical data of the rat testis were studied between the 1st day and 1.5 year of life under physiological conditions and after intraperitoneal injection of a single dose of cadmium chloride (1.5 mg CdCl2/kg body weight). Under physiological conditions three main period of development occur: between the 1st and 30th day of life, between the 30th and 45th day of life, and after the 45th day of life. The greatest changes after the injection of cadmium were noticed in the period between the 15th and 30th day of life. The periphery of the organ was the first to suffer severe damage.
