Identification and characterization of the two-enzyme system catalyzing oxidation of EDTA in the EDTA-degrading bacterial strain DSM 9103.

In a gram-negative isolate (DSM 9103) able to grow with EDTA as the sole source of carbon, nitrogen, and energy, the first two steps of the catabolic pathway for EDTA were elucidated. They consisted of the sequential oxidative removal of two acetyl groups, resulting in the formation of glyoxylate. An enzyme complex that catalyzes the removal of two acetyl groups was purified and characterized. In the reaction, ethylenediaminetriacetate (ED3A) was formed as an intermediate and N,N'-ethylenediaminediacetate was the end product. The enzyme complex consisted of two components: component A' (cA'), most likely a monooxygenase, which catalyzes the cleavage of EDTA and ED3A while consuming oxygen and reduced flavin mononucleotide (FMN)-H2, and component B' (cB'), an NADH2:FMN oxidoreductase that provides FMNH2 for cA'. cB' could be replaced by other NADH2:FMN oxidoreductases such as component B of the nitrilotriacetate monooxygenase or the NADH2:FMN oxidoreductase from Photobacterium fischeri. The EDTA-oxidizing enzyme complex accepted EDTA as a substrate only when it was complexed with Mg2+, Zn2+, Mn2+, Co2+, or Cu2+. Moreover, the enzyme complex catalyzed the removal of acetyl groups from several other aminopolycarboxylic acids that possess three or more acetyl groups.

Quantification of Bcr-Abl transcripts in chronic myelogenous leukemia (CML) using standardized, internally controlled, competitive differential PCR (CD-PCR).

The quantification of Bcr-Abl transcript numbers in chronic myelogenous leukemia (CML) patients described here uses simultaneous competitive PCR amplification of the target gene (Bcr-Abl) and a reference gene (porphobilinogen deaminase; Pbgd) together with a single composite competitor molecule for both targets based on heterologous sequences. Using this technique, Bcr-Abl transcript numbers could be reproducibly determined even in clinical samples known to harbour poor quality RNA.

[Light scattering study of the cornea in contact lens patients. In vivo studies using confocal slit scanning microscopy].

For the first time, we present measurements of the scattered light intensity of the human cornea to determine the functional state of the corneal endothelium. In 94 contact lens wearers endothelial alterations were measured by means of a newly developed confocal slit scanning video microscope and correlated with photometrically recorded stromal scattered light intensity. Our control group consisted of 26 persons without corneal pathology. It was shown that in persons wearing contact lenses the endothelial cell density was significantly below that of the control group and endothelial cell polymegatism (coefficient of variability of cell size) was significantly greater. Further, in contact lens wearers the level of scattered light intensity was significantly higher than in the control group, and the difference increased with the wearing time. In addition, the variation of the endothelial cell size correlated with the increased scattered light within the stroma, only a low correlation was found between the endothelial cell density and the scattered light intensity, however. From our results it can be assumed that a corneal endothelium with marked polymegatism has a reduced hydration function. Thus, decreased endothelial function can be determined very early and non-invasively by measuring the corneal scattered light intensity. In addition, the scattered light intensity gives topographical information about corneal swelling.

Exposure to polychlorinated dioxins and furans (PCDD/F) and mortality in a cohort of workers from a herbicide-producing plant in Hamburg, Federal Republic of Germany.

The relation between mortality (all cause; cancer; cardiovascular diseases (International Classification of Diseases, Ninth Revision (ICD-9), codes 390-459); ischemic heart diseases (ICD-9 codes 410-414)) and exposure to polychlorinated dibenzo-p-dioxins and -furans (PCDD/F) was investigated in a retrospective cohort study. The cohort consisted of 1,189 male workers in a chemical plant in Hamburg, Federal Republic of Germany, who had produced phenoxy herbicides, chlorophenols, and other herbicides and insecticides known to be contaminated with 2,3,7,8-tetrachlorodibenzo-p-dioxin and other, higher chlorinated dioxins and furans. The authors reported previously on cancer mortality in this cohort for the follow-up period 1952-1989. The current study covers the years 1952-1992 and investigates the relation of PCDD/F exposure to mortality using a quantitative estimate of PCDD/F exposure for the whole cohort derived from blood and adipose tissue levels measured in a subgroup (n = 190). Quintiles and deciles of these estimates served as dose parameters in the estimation of relative risks (RRs), using year-of-birth stratified Cox regression. An unexposed cohort of gas workers served as an external reference group. The total mortality was elevated in all dose groups. The highest relative risk was observed for the highest 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) decile (RR = 2.43, 95% confidence interval (CI) 1.80 to 3.29). Cancer mortality and mortality due to ischemic heart diseases showed a dose-dependent relation with TCDD and all PCDD/F combined. The highest relative risks for cancer (RR = 3.30, 95% CI 2.05 to 5.31) and ischemic heart diseases (RR = 2.48, 95% CI 1.32 to 4.66) were observed in the highest PCDD/F exposure group. The pattern of effects and tests for trend were similar when the lowest exposure group within the chemical worker cohort served as the reference, but the relative risks were smaller and the confidence intervals were larger. Potential confounding exposures complicate the interpretation of the internal comparison. These findings indicate a strong dose-dependent relation between mortality due to cancer or ischemic heart diseases and exposure to polychlorinated dioxins and furans.

Somatic mutations detected by mini- and microsatellite DNA markers reveal clonal intratumor heterogeneity in gastrointestinal cancers.

We investigated clonal intratumor heterogeneity by comparing different areas of each tumor in 20 gastrointestinal cancers from female patients (1 esophageal cancer, 5 stomach cancers, and 14 colorectal cancers). In all 19 cases informative for X-inactivation analysis with the M27 beta and/or the phosphoglycerate kinase probes, the tumors were clonal. Separate areas from a given tumor showed identical X-inactivation patterns, providing evidence for its single-cell origin. Of 20 cancers, 11 showed p53 gene mutations (base pair insertions, point mutations, and one case of a base pair deletion) in exons 5-8. A particular p53 gene mutation was identical in all tumor areas investigated per case. The minisatellite probes detected loss of heterozygosity or new mutant alleles at 1p33, 1q21, 5q35, 17p13, or 18q21. In seven cases mutations at particular loci were restricted to one or two areas per tumor, while in another seven cases they were common to all tumor areas. Loss of heterozygosity or new alleles detected at the microsatellite loci D2S123, D3S1611, D5S107, D17S261, or D18S34 [(CA)n repeats] were common to all tumor areas in 7 of 19 cases. In another seven cases, however, microsatellite mutations at these loci were restricted to one to three areas per tumor. Tracing clonal intratumor heterogeneity would permit one to study the hierarchy of mutational events in cancers where no premalignant lesions can be harvested. Most important, our study indicates that clonal intratumor heterogeneity might lead to sampling errors in the molecular diagnosis of cancer biopsy specimens when using mini- or microsatellite markers.

Posttranscriptional stabilization underlies p53-independent induction of p21WAF1/CIP1/SDI1 in differentiating human leukemic cells.

p21WAF/CIP1/SDI1 is a recently identified gene expressed in cells harboring wild-type but not mutant p53 gene. It encodes a nuclear protein of 21 kD which inhibits cyclin-dependent kinase activity. Constitutive p21WAF1/CIP1/SDI1 mRNA expression was detected in neoplastic cells from patients with various hematological malignancies as well as in normal bone marrow mononuclear cells and in myeloid and lymphoid cell lines independent of their p53 status. Induced differentiation of the p53-deficient promyelocytic HL-60 cells along the monocytic lineage by phorbol ester or 1a,25 dihydroxyvitamin D3 resulted in a marked increase of both p21WAF1/CIP1/SDI1 mRNA and protein expression due to enhanced mRNA stability. Differentiation towards the granulocytic lineage by all-trans retinoic acid or dimethylsulfoxide failed to produce this effect. p21WAF1/CIP1/SDI1 is an immediate early gene since its upregulation occurred independently of de novo protein synthesis. The induction of p21WAF1/CIP1/SDI1 expression and its regulation in p53-deficient differentiating leukemic cells support the idea of an additional, p53-independent role of p21WAF1/CIP1/SDI1 in human hematopoiesis.

[Corneal changes and corneal healing after keratomileusis in situ. In vivo studies using confocal slit-scanning microscopy].

So far, no investigations of micromorphological changes in the human cornea after in situ keratomileusis have been published. In 10 eyes of five patients who had undergone in situ keratomileusis, wound healing processes were followed up with a newly developed confocal slit-scanning video-microscope. Post-operatively, alterations in the corneal endothelium and in the corneal stroma were found. Wide streaky distortions were observed within the stroma above and below the lamellar incision during the early days immediately after surgery. These distortions were probably induced by shearing stresses of the oscillating blade of the microkeratome. Small scattering centers were seen at the level of the lamellar incision, which are thought to reflect a very slight roughness of the interface. In the immediate postoperative period the corneal endothelium showed reversible inhomogeneitis similar to blebs after short-term contact lens wearing, which are thought to be due to a hypoxia during the surgery. The endothelial alterations and the streaky distortions within the stroma disappeared within 9 days after the surgery whereas the scattering centers regressed much more slowly and could still be observed 60 days after the surgery. The total amount of scattered light of the cornea, however, was not increased except in one patient. The results show that outside the level of the lamellar incision, in situ keratomileusis leads to only short-term alterations of the corneal micromorphology, and that confocal in vivo slit-scanning video-microscopy allows thorough examination of these alterations.

Clonality and X-inactivation patterns in hematopoietic cell populations detected by the highly informative M27 beta DNA probe.

About 80% to 90% of females are informative for X-inactivation/methylation analysis with the probe M27 beta, which would therefore seem attractive in assessing clonality in hematologic cell populations. Eighteen acute lymphoid or myeloid leukemias, three chronic lymphocytic leukemias, and three chronic myelogenous leukemias as well as 12 malignant non-Hodgkin's lymphomas mostly showed extremely skewed clonal X inactivation (median allelic cleavage ratio [ACR] of unmethylated/inactive M27 beta alleles was 50, range 1 to 100) or hypermethylation of both alleles. Two lymphomas showed random M27 beta X inactivation but clonal antigen-receptor gene rearrangements. In normal peripheral blood leukocytes from 105 healthy females aged 2 to 96 years, the median ACR was 2 (range 1 to 100). Thus, it was significantly lower than in the leukemias (P = .0001, Mann-Whitney test), but extremely skewed patterns (ACR > 10.8, ie, > 80th percentile) were seen not only in the leukemias but also in 21/105 samples (20%) of normal leukocytes, and significantly more frequent in a population of elderly women (aged 75 to 96 years) compared with healthy children (aged 2 to 8 years) and younger women (aged 20 to 58 years) (P = .00125; chi 2 test). We conclude that in a population of cells derived from the hematopoietic system where clonality is uncertain, skewed M27 beta patterns are not reliable indicators for the presence of a clonal neoplastic disorder. The basis for severe X-inactivation skewing is unclear at present, but this finding raises interesting questions regarding the composition of the hematopoietic stem cell pool.

Structurally divergent histone H1 variants in chromosomes containing highly condensed interphase chromatin.

Condensed and late-replicating interphase chromatin in the Dipertan insect Chironomus contains a divergent type of histone H1 with an inserted KAP-KAP repeat that is conserved in single H1 variants of Caenorhabditis elegans and Volvox carteri. H1 peptides comprising the insertion interact specifically with DNA. The Chironomid Glyptotendipes exhibits a corresponding correlation between the presence of condensed chromosome sections and the appearance of a divergent H1 subtype. The centromere regions and other sections of Glyptotendipes barbipes chromosomes are inaccessible to immunodecoration by anti-H2B and anti-H1 antibodies one of which is known to recognize nine different epitopes in all domains of the H1 molecule. Microelectrophoresis of the histones from manually isolated unfixed centromeres revealed the presence of H1 and core histones. H1 genes of G. barpipes were sequenced and found to belong to two groups. H1 II and H1 III are rather similar but differ remarkably from H1 I. About 30% of the deduced amino acid residues were found to be unique to H1 I. Most conspicuous is the insertion, SPAKSPGR, in H1 I that is lacking in H1 II and H1 III and at its position gives rise to the sequence repeat SPAKSPAKSPGR. The homologous H1 I gene in Glyptotendipes salinus encodes the very similar repeat TPAKSPAKSPGR. Both sequences are structurally related to the KAPKAP repeat in H1 I-1 specific for condensed chromosome sites in Chironomus and to the SPKKSPKK repeat in sea urchin sperm H1, lie at almost the same distance from the central globular domain, and could interact with linker DNA in packaging condensed chromatin.

Morbidity and mortality following abdominoperineal resection for rectal adenocarcinoma.

From 1979 through 1986, 56 patients, 38 males (68%) and 18 females (32%), underwent an abdominoperineal resection with a wide perineal dissection for primary adenocarcinoma of the rectum. The open perineal wounds required a mean time of 6 months to completely heal. Estimated blood loss by the surgeon ranged from 200 ml to 4200 ml with a mean of 1112 ml and a median of 1000 ml. Twenty-six patients (46%) developed postoperative or intraoperative complications. Nineteen patients (34%) underwent incidental surgery with no associated morbidity. The most commonly occurring complication was impotence in 69% of male patients. The next most common complication was bladder dysfunction with urinary retention in 14 patients (25%) and urinary incontinence in five patients (9%). There were two postoperative deaths (4%). Although the morbidity was significant, greater than 90% of the complications were treatable and resolved without major sequellae.

Cancer mortality among workers in chemical plant contaminated with dioxin.

Dioxin (2,3,7,8-tetrachlorodibenzo-p-dioxin or TCDD) can arise as a contaminant in the production of herbicides. It causes chloracne in those exposed to it but its human carcinogenicity has been a matter of dispute. We report here a mortality follow-up of 1583 workers (1184 men, 399 women) employed in a chemical plant in Germany that produced herbicides, including processes contaminated with TCDD. Production of TCDD was reduced from 1954 after an outbreak of chloracne. Vital status up to 1989 was determined for 97.1% of workers hired between 1952 and 1984, and 367 deaths (313 men, 54 women) were recorded. A malignant neoplasm was the underlying cause of death in 93 men and 20 women. Standardised mortality ratios (SMR) were calculated with, as references, national mortality statistics for West Germany and deaths in a cohort of male gas workers; for total cancer mortality they were 1.24 (95% confidence interval 1.00-1.52) and 1.39 (1.10-1.75), respectively, among men. Cancer mortality was increased among men with 20 or more years of employment (SMR = 1.87 [Germany] and 1.82 [gas workers]) and among men who began employment before 1955 (SMR = 1.61 and 1.87). The group with suspected highest exposure to TCDD had SMRs of 1.42 and 1.78. Only 7% of cohort women worked in the high exposure locations in the plant, compared with 39.6% of men, and no increased risk of cancer mortality was observed among women; but breast cancer mortality was raised (SRM = 2.15). These results, together with a US occupational study and a German investigation of accidental exposure, support the hypothesis that TCDD is a human carcinogen.

Studies on the evaluation of feed protein for ruminants. 1. Passage of nitrogen fractions to the duodenum of dairy cows fed different protein and carbohydrate sources.

The duodenal passages of non-ammonia nitrogen (NAN), amino acid N (AA-N) and microbial N (MN) were measured in seven duodenal fistulated dairy cows (6130 kg FCM/305 d) during lactation receiving 30 different rations similar to those used in dairy practice. The rations consisted of roughage (protein-rich silages, hay) and concentrate mixture (corn, barley, oat) in a ratio of 60:40 on a DM basis or roughage/concentrate mixture/fodder beets in the proportions 60:20:20 or 60:0:40 respectively. N supplements consisted of soya bean meal and peas meal (untreated or treated with formaldehyde in each case), rapeseed meal, fish meal and urea. DM intake varied between 9.6 and 19.1 kg/d, the crude fibre content between 166 and 270 g/kg DM, the crude protein (CP) content between 111 and 184 g/kg DM, the AA content (N basis) of the CP between 51 and 82%, the unfermentable CP content of the CP between 23 and 49% and the organic matter digestibility between 70 and 79%. The duodenal passage of NAN was 24.2 +/- 2.7 g/kg DM intake, 34.9 +/- 4.1 g/kg apparently digestible organic matter (in total tract, DOM), 43.3 +/- 5.5 g/kg apparently digestible carbohydrates or 4.2 +/- 0.5 g/MJ net energy fat, (y +/- s, n = 90). The value measured for AA-N was 16.8 +/- 2.9 g/kg DM intake, and that for RNA-labelled microbial CP was 146 +/- 26 g/kg DOM. NAN passage (g/kg DM intake) correlated more closely with the duodenal digesta flow rate (DFR) (kg digesta/kg DM intake) as an intrinsic animal factor (r = 0.78) and with the rumen content dilution rate (RDR) (passage of microbial-free organic matter at duodenum/kg BW0.75.h) as a ration dependent factor (r = 0.64) than with the UDP fraction (% of ration CP) (r = 0.50) or other ration parameters. It was concluded that the specific effect of protein concentrates in mixed rations on duodenal NAN yields in lactating dairy cows is lower than hitherto assumed. The duodenal NAN yield of a single feedstuff or ration (g/kg DM) in cows is dependent on animal and feeding factors. The DFR could represents a suitable target for breeding activities.
