A tetrazolium method for staining peroxidase labels in blotting assays.

A sensitive staining method of horseradish peroxidase-labeled immunoglobulins on nitrocellulose membrane was established by employing a reaction chain leading to formazan formation with phenol as a substrate of peroxidase and NADH as a hydrogen donor to reduce nitro blue tetrazolium. Higher concentrations of NADH relative to phenol were necessary to increase the intensity of staining and to ensure a wide dose-response range of color production with respect to the applied enzyme activities. By an optimized tetrazolium method in combination with antibody-affinity blotting, as low as 4 ng/ml alpha-fetoprotein was detected and 3-4-fold greater color intensities in a working assay range as compared with those of existing methods were obtained. The present technique of peroxidase staining may prove to have a wide application for the enzyme immunoassay using blotting modalities.

Pure red cell aplasia in association with malignant histiocytosis.

The authors report a case of malignant histiocytosis complicated with pure red cell aplasia. Erythrophagocytic histiocytes were found only rarely and no histiocyte ingested erythroblasts. Neither serum immunoglobulin G nor peripheral blood T-cells have any inhibitory activity to erythroid colony growth from allogeneic bone marrow. Pure red cell aplasia is an unusual complication in malignant histiocytosis.

T cell-mediated inhibition of haematopoiesis in aplastic anaemia: serial assay of inhibitory activities of T cells to autologous CFU-E during immunosuppressive therapy.

T cell-mediated inhibition of autologous erythroid colony formation was found in two patients with aplastic anaemia. Each patient was treated separately with ALG and methylprednisolone. Peripheral blood T cells were cryopreserved serially during the course of the disease. The inhibitory activity of T cells was assayed after remission using autologous bone marrow. The inhibitory activity of T cells was lost following the treatment and preceded haematopoietic recovery.

[A statistical approach to Yatabe-Guilford personality inventory and manifest anxiety scale of the elderly].

In nursing, if we know an elderly person's mental condition, and that which does not make him/her too tired, this method can be very useful. Using this method we predicted the MAS score from scores of Y-G scales. We obtained the MAS score results by the following formula: MAS score not equal to 0.31 X Lack of objectivity +0.41 X Inferiority feelings -0.15 X Ascendance +0.34 X Y-G Nervousness +0.30 X Y-G Depression +9.54.
