[The nature of virus strains isolated from patients with poliomyelitis in the Byelorussian SSR 1967-1985].

Oligonucleotide mapping of the RNA genome was applied for analysis of poliovirus clinical isolates in Byelorussia in 1967-1985. All the maps have been clearly shown to resemble the reference maps of the corresponding vaccine strains. Examination of the oligonucleotide maps of poliovirus isolates provides a measure of distribution of some of the mutations over the viral genome.

[Antigenic polymorphism in amyotrophic lateral sclerosis].

HLA-antigen distribution in 93 patients with amyotrophic lateral sclerosis did not differ from that in the control group. Yet, there were significant differences in antigen levels both between different variants of the disease and as compared to the control values.

[Epidemiological research in amyotrophic leukospongiosis].

The epidemiological aspects of amyotrophic leukospongiosis (AL), a slow viral infection of the central nervous system leading to the fatal outcome in 2-4 years, have been studied. As a rule, this disease is observed in the inhabitants of rural areas or in town dwellers born in rural areas and having spent there a considerable part of their life. AL occurs in persons of middle and older age; young people under 19 years and old people over 68 years of age are not affected by this infection. In contrast to amyotrophic lateral sclerosis and the Jakob-Creutzfeldt disease, AL is characteristic for persons in the phase of hormonal activity. The disease starts mostly in autumn and winter; this regularity is especially pronounced in women. The morbidity level (according to the average annual data) is at present 0.3 per million of population. An increased morbidity rate is characteristic of the family and group type of the epidemic process. This higher morbidity rate, by one order higher than that observed in the sporadic type of morbidity, is caused by the gradual formation of "genetic isolates".

[The ATP depletion process of human erythrocytes studied by freeze fracturing].

By the freeze-fracture method it is shown that metabolic depletion of erythrocytes affects three levels of cell organization: the microstructural (erythrocyte form), microstructural (micro-relief of erythrocyte surface) and ultrastructural (ultrastructural state of erythrocyte plasma membranes) ones. As it is established, the size of spikes on the echinocyte surface and that of membrane vesicles budding from a cell coincide with each other. The structural modification of the membrane precedes the stage of erythrocyte crenation. The following model of vesicle budding process is suggested: reduction of ATP level and dephosphorylation of actin-spectrin network--structural modification of the protein and lipid membrane phases with the formation of regions disconnected from the spectrin framework--protrusion of these anomalous regions in the form of spikes--budding of spikes as spherical vesicles.

[Development of laboratory diagnostic methods for slow infections of the human central nervous system caused by nonclassical viruses].

The theoretical foundations of the laboratory diagnosis of Creutzfeldt-Jakob disease and amyotrophic leukospongiosis in living patients are discussed and practical diagnostic methods are proposed on the basis of the study of the biological properties of nonclassical viruses and the pathogenesis of slow infections of the central nervous system. The use of the retrobulbar method of infecting susceptible animals has permitted, besides a considerable decrease in the incubation period in modeling slow infections, the improvement of the in vivo biological test. Thus, as the result of the multiplication of non-classical viruses in the visual nerve and the retina, experimental animals develop retinopathy which can be detected 1.5-2.5 weeks after the animals are injected with the spinal fluid or blood taken from the patient under examination. To diagnose amyotrophic leukospongiosis, the in vitro biological test, based on the capacity of the causative agent for inducing a considerable (2- to 5-fold) increase in the mitotic activity of HEp-2 cells, has been developed. The comparative studies have shown the expediency of using the method for the detection of antibodies to nerve-fiber proteins as an additional test for the differential diagnosis of slow infections of the central nervous system.

[Effect of adenine nucleotides on the activator function of streptokinase].

The addition of ATP or 3,5-AMP (but not UTP, GTP, CTP, AMP, 2,3-AMP, ADP, inorganic pyrophosphate) at a final concentration of 10(-1) M into streptokinase solution, pH 7.0 or 9.5, causes a dramatic inhibition of streptokinase-induced fibrinolysis. The specificity of ATP effect is fully lost at pH 3.0, when all nucleotides completely inhibit the activating function of streptokinase. Ribose-5-phosphate causes a similar effect at pH 3.0. The character of nucleotide action on the activating function of streptokinase considerably differs from their influence on proteolytic reactions.

[Action of streptokinase on the hemostatic indices of rabbits with a toxic lesion of the liver due to carbon tetrachloride].

The rabbits with CCl4-induced hepatic failure have revealed changes in hemostasis responses to streptokinase administration. The main distinction of hepatic dystrophy was the depression of plasma fibrinolytic activity accompanying the decrease in fibrinogen and antiplasmin concentrations. Streptokinase administration to rabbits with productive inflammatory liver disorders produced changes in hemostasis identical to those observed in intact rabbits, fibrinogen levels, however, remained unchanged. The common feature of all the toxic liver disorders is the increase of antithrombin III levels after streptokinase administration, whereas the antithrombin levels in the control animals were decreased.

[Biochemical and immunological changes in guinea pigs with experimentally reproduced amyotrophic leukospongiosis].

Biochemical blood serum tests at different stages of amyotrophic leukospongiosis have shown differences in lactate and pyruvate levels as well as in lactate dehydrogenase activity, indicative of the increased oxidative exchange in sick guinea-pigs. It is suggested that intensified glycolysis is a compensatory-adaptive reaction in response to hypoxia due to respiratory disorders (spinal type) and degeneration and death of motoneurons. Leukospongiosis was accompanied by the decline in the complement level in the blood serum and production of antibodies to nervous fiber proteins.

[Isolation of a nonclassical virus and a study of its etiological role in human amyotrophic leukospongiosis].

The study of the brain of a patients with amyotrophic leukospongiosis and the brain of squirrel monkeys with the experimentally induced disease has resulted in the isolation of an agent which, in accordance with its physicochemical and biological properties, can be classified with unconventional viruses. The results of the experimental reproduction of the disease in laboratory animals have confirmed its etiological role in the development of the disease.

[Experimental amyotrophic leukospongiosis in guinea pigs after retrobulbar infection].

The results of modelling of amyotrophic leukospongiosis, a new form of slow infection of the human central nervous system, on guinea-pigs are reported. The animals were injected retrobalbulary with the virus-containing suspension. 1.5-2.5 weeks after the injection 70% of animals revealed signs of retinopathy during ophthalmoscopy. Two months later 90% of animals died, 40% of them exhibiting manifestations of the infection. Experimental amyotrophic leukospongiosis was histologically confirmed in all the dead animals. This method of modelling made it possible to shorten the incubation period to 1.0-2.0 months, while in intracerebral and intramuscular ways of contamination it was 3.5-8.2 and 5.3-11.1 months, respectively. The results evidence the involvement of the peripheral visual analyzer in the pathogenesis of experimental leukospongiosis at the early stages of its development.
