[Prevention of viral hepatitis in children with concentrated human leukocyte interferon].

Concentrated human leukocytic interferon, introduced electrophoretically in small doses for preventive purposes, reduces morbidity rate in hepatitis foci 6 times, probably due to an increase in the production of endogenous interferon. Interferon is introduced once in 7-10 days, taking into account the minimal time of the preservation of the preparation in the musculocutaneous depot. The application of a rubber bandage and the creation of the zone of congestive hyperemia leads to deeper penetration of interferon and to the increased therapeutic effectiveness of the preparation. The absence of significant differences in a number of immunological characteristics may be attributed to the use of interferon for the prophylaxis of healthy children having had contacts with viral hepatitis patients rather than children, actually having the disease.

[Effect of interferon type I on the in vivo persistence of Salmonella typhimurium].

The influence of type I interferon on the persistence of S. typhimurium in the body of mice has been studied. The injection of the preparation of interferon has been shown to be conductive to the survival of the animals and to reduce the time of Salmonella persistence in the body. The injection of interferon enhances the phagocytic activity of macrophages in the peritoneal exudate of mice.

[Treatment methods in chronic bronchitis].

Patients with chronic bronchitis at exacerbation and in remission were treated by immunocorrective agents. A study was made of the immune status, interferon synthesizing capacity of blood leukocytes, a clinical course of disease and their change resulting from immunocorrective therapy provided at different stages of chronic bronchitis. The authors established disorders in the immune status, a decrease in interferon synthesizing capacity of blood leukocytes which were more noticeable at exacerbation. The use of immunocorrective therapy raised the production of interferon by the patients' blood leukocytes, improved ratios between lymphocyte populations and subpopulations, considerably improved a clinical course of disease, prolonged remission, facilitated a course of exacerbations and decreased resistance to anti-bacterial therapy.

[Antitoxic activity of interferon preparations].

The antitoxic activity of leucocytic injection interferon I and immune interferon was shown in experimental erythrocyte hemolysis in the presence of staphylococcus alpha-toxin. The antitoxic effect was directly proportional to the interferon concentration in the medium and inversely proportional to the toxin concentration. Neutralization of the antiviral activity of leucocytic interferon did not lower its antitoxic effect. The highly purified and concentrated preparation of leucocytic injection interferon I and recombinant interferon had no antitoxic effect.

Studies of proteins soluble in acidic chloroform-methanol isolated from crude human leukocyte interferon preparations.

Acidic chloroform-methanol soluble proteins possessing hydrophobic properties and capable of inhibiting in vitro transcriptase activity of influenza virus RNP were detected in native and partially purified human leukocyte interferon (IFN) preparations. Purification of IFN resulted in the removal of at least a portion of such proteins; however, no proteins have been found in highly-purified IFN preparations.

[Inactivation of the inductor virus (Newcastle disease virus) during the isolation of porcine leukocyte interferon].

The use of the routine method for inactivation of the inductor virus by acidification of interferon to pH 2.0 resulted in a significant decrease in the antiviral activity of pig leukocytic interferon, since the preparation was highly sensitive to changes in the pH values. The use of 0.1 per cent solution of formalin provided complete inactivation of the virus. The antiviral activity of interferon treated with formalin was on an average 5 times higher than that of the preparation incubated at pH 2.5-2.6. Precipitation of pig interferon with polyethylene glycol promoted both increasing of the titers of the antiviral activity of the preparation and elimination of the formalin residues from it. Interferon prepared with this procedure was not toxic in tissue cultures, had no side effects when applied to the eye mucosa and was absolutely harmless when administered to animals. It was shown that inactivation of the inductor virus with formalin was in principle possible in human leukocytic interferon.

[Interferon inhibition of the development of a staphylococcal infection].

Administration of interferon to mice with staphylococcal infection promoted elimination of the bacteria from their organs, influenced the immunological indices, normalized delayed type hypersensitivity, recovered the capacity of the spleen cells to produce interferon and increased the phagocytic activity of the peritoneal macrophages against the staphylococci. Addition of alpha-interferon to antibiotic therapy of patients with purulent septic infections of staphylococcal etiology markedly increased its efficiency. Improvement of the clinical picture of the disease, normalization of the general biochemical indices of the blood and elimination of the staphylococci from the blood and inflammation foci were observed.

Antiviral activity of lambda-, beta-, and gamma-interferons in the presence of theophylline: involvement of 2',5'-oligo(A) synthetase.

Theophylline, an inhibitor of cAMP phosphodiesterase, induces in human ovary carcinoma cells (CaOv) a 2-2.5-fold elevation of intracellular cAMP. This rise in the cAMP level is followed by an increase of the activity of 2',5'-oligo(A) synthetase in CaOv cells -insignificant (1.5-fold) after 16 hr incubation, and substantial (3.7-fold) after 30 hr incubation, as well as the development of antiviral resistance. Once CaOv cells have been incubated with the mixtures containing theophylline (2 mM) and lambda-, beta-, and gamma-interferon preparations (0.5-13 IU/ml), the total antiviral effect of the mixtures exceeds that generated by interferon or theophylline separately; the action of the above agents being additive. These data agree with the previously obtained results and support the suggestion that cAMP phosphodiesterase inhibitors partially mimic the antiviral action of interferon.

[Ratio of the antiproliferative action of interferon to the degree of purity of the preparation].

The antiproliferative (AP) action of human leukocytic interferon (HLI) of varying degrees of purification was studied and compared in cultures of human cells differing in interferon sensitivity. The concentrated and purified preparations of HLI suppressed the incorporation of 3H-thymidine and mitotic activities of the cultures sensitive to HLI of the cells, affecting, but insignificantly, the proliferation of the resistant cell culture. Poorly purified HLI suppressed the proliferation of both cell types. This indicates that native HLI contains impurities that have an AP action and exerts no antiviral activity.
