The uterus of six healthy multiparous women at term was mechanically stretched by a rubber catheter and balloon. Apparent labor was inaugurated in all cases within 5 hours and increased progressively with time. Advanced cervical softening and dilatation were also evident after the stretch treatment. Significant increases in the levels of 13,14-dihydro-15-keto-prostaglandin F2 alpha (PGFM) were observed with the progress of treatment (P less than 0.01). Plasma estrogens and progesterone levels did not change significantly during the treatment (P greater than 0.05). Stretching and/or resulting uterine contractions appear to induce the secretion of prostaglandin F2 alpha (PGF) from the organ, which in turn seems to be involved in both cervical softening, and the onset and progress of labor, under stable conditions of plasma estrogens and progesterone.

Prognosis of differentiated thyroid carcinoma is favorable unless distant metastasis develops. We have experienced 70 cases of differentiated thyroid carcinoma with pulmonary metastases and analysed the outcome of treatment of patients, with special reference to the extent of thyroid resection and postoperative ablative RI therapy. The following results were obtained: Of the 42 survivors 22 (52.4%) received total thyroidectomy, whereas 23 of 28 expired patients underwent less-than-total thyroidectomy. A total of 42 patients underwent radioiodine treatment following thyroid resection. 17 out of 18 patients who had received total thyroidectomy survived. On the other hand, only 11 out of 24 patients who had received less-than-total thyroidectomy survived. In the non-RI treatment group, prognoses were unfavorable in both total and less-than-total thyroidectomy groups. From these results, it is emphasized that ablative doses of RI should be given to patients with pulmonary metastases of differentiated thyroid carcinoma after total thyroidectomy.
The ability of high-dose intravenous gammaglobulin (IVGG) to prevent the coronary artery lesion of Kawasaki disease has been studied in a multicentre controlled trial of IVGG plus aspirin versus aspirin alone, aspirin being the conventional treatment for Kawasaki disease. Patients were allocated at random to aspirin (45 cases) or IVGG (40 cases), there being no significant intergroup differences in age, sex ratio, duration of disease until the start of treatment, or severity. The development of coronary artery dilatation was monitored by two-dimensional echocardiography. Within 29 days of the onset of the disease, this lesion had developed in 19 cases (42%) in the aspirin group and in 6 cases (15%) in the IVGG group. There were no new instances of this lesion: in the period 30-60 days coronary artery dilatation persisted in 14 and 3 cases, respectively. In patients found to have echocardiographic abnormalities selective coronary arteriography was done 30-60 days after onset of Kawasaki disease and the lesion was confirmed in 1 of the 6 cases in the IVGG group and in 11 of the 19 controls. High-dose IVGG seems to reduce the frequency of coronary artery abnormalities in patients with Kawasaki disease.

Wastewater collected from oil-water separating tanks of ten gasoline stations for a year was fractionated into diethyl ether-soluble neutral, acidic, and basic fractions. Mutagenicity of these fractions was measured with Salmonella typhimurium strains TA98 and TA100 in the presence or absence of S9 mix. The neutral fractions showed high mutagenicity in the absence of S9 mix. Each neutral fraction was subjected to high-performance liquid chromatography (HPLC) and fractionated. A 1-nitropyrene(1-NP)-corresponding fraction was collected and analyzed by gas chromatography-mass spectrometry (GC-MS) and HPLC to prove that wastewater contains 1-NP and to quantitate 1-NP in wastewater. GC-MS patterns showed the following molecular and fragment ion peaks of 1-NP: 247, 217, 201, and 189. The amount of 1-NP in 36 samples of wastewater was 4.2-25,600 ng per liter of wastewater, and 1-NP accounted for 0.3-58.5% of the total mutagenicity of the neutral fractions. The other 19 samples of wastewater did not contain any detectable 1-NP. The mutagenicity of wastewater may be due to water from car washing and contamination by used crankcase oil. A Soxhlet extract of crankcase oil used in a gasoline was fractionated into three fractions as above. Mutagenicity was measured with strains TA98, TA100, TA98NR, and TA98/1,8-DNP6 in the absence or presence of S9 mix. The neutral fraction showed the highest mutagenicity with strain TA98 in the absence of S9 mix, and its mutagenicity was decreased in strains TA98NR and TA98/1,8-DNP6. The latter result indicates that the used engine-oil contained 1-NP and dinitropyrenes. Actually, the amounts of 1-NP and 1,6-diNP in used crankcase oil were 138 and 2.0 ng per ml of oil, respectively, and these concentrations accounted for 0.45 and 2.7%, respectively, of the total mutagenicity of the neutral fraction with strain TA98 in the absence of S9 mix. Moreover, the concentrations of 1-NP and 1,6-diNP in used crankcase oil of a diesel engine were 349 and 31 ng per ml of oil, respectively, accounting for 0.9 and 12%, respectively, of the total mutagenicity of the neutral fraction in the same assay system.
Macromomycin (MCR), an unique membrane-reactive anticancer antibiotic, was incubated with murine monoclonal anti-HLA IgG1 antibody (H-1) in the presence of carbodiimide. The resulting mixture was fractionated with a Sephadex G-200 column. The first and second fractions were shown to contain MCR-(H-1) conjugate by the elution profile, as well as by the Sarcina lutea growth inhibition assay and Ouchterlony double-diffusion method. A membrane immunofluorescence test with anti-MCR and anti-mouse IgG antibodies demonstrated specific localization of MCR-(H-1) on the surface of HLA-bearing NALL -1 cells. MCR-(H-1) inhibited the growth of HLA-lacking NS-1 cells statistically less effectively than MCR alone (p less than 0.01). On the other hand, the conjugate and free MCR equally inhibited the growth and 3H-TdR incorporation of HLA-bearing NALL -1 cells. These results indicate that the antibody-bound MCR retained both MCR and antibody activities, and thus exerted antibody-targeting MCR cytotoxicity in vitro.
Methotrexate (MTX), a folate antagonist and anticancer agent, was linked covalently to murine monoclonal anti-HLA IgG1 antibody (H-1) with the use of dextran T-40 as a multivalent carrier. As determined spectrophotometrically, the conjugate was composed of 9.20 mol MTX per 1 mol antibody. Of the substituted MTX, 46.3% (4.30 mol MTX per 1 mol antibody) exhibited inhibitory activity on dihydrofolate reductase. The MTX conjugated to H-1 [MTX-(H-l)] showed significantly stronger cytotoxicity against HLA-bearing cells than cells lacking HLA (p less than 0.001) in cultivation after a 2-hour exposure to the drugs. In the same experiment, free MTX showed equivalent cytotoxicity to cells both bearing and lacking HLA. When cells were cultivated for 3 days with the drugs, MTX-(H-l) also showed significantly stronger cytotoxicity than free MTX or MTX-control IgG conjugate against HLA-bearing cells (p less than 0.001), and the MTX-(H-l), MTX-control IgG, and free MTX showed equivalent cytotoxicity to cells lacking HLA. Our results indicate that the MTX-(H-l) conjugate binds to the cell surface antigen by its antibody action and exerts greater MTX cytotoxicity than free MTX in vitro.
In tests on the carcinogenicity of 1,6-dinitropyrene [(1,6-DNP) CAS: 42397-64-8] and 1-nitropyrene [(1-NP) CAS: 5522-43-0], 0.1 mg of each compound was inoculated sc into BALB/c mice once a week for 20 weeks. In the group given injections of 1,6-DNP the first tumor appeared on day 112, and 10 of the 20 mice developed tumors at the injection site by 45 weeks after the first injection. However, no tumors were induced in any of the mice that received injections of 1-NP. All of the induced tumors were transplantable for more than five generations in male BALB/c mice. Most of the tumors showed the characteristic histologic features of malignant fibrous histiocytoma.
Two to four stems of Laminaria were inserted into the cervixes of seven healthy women at second trimester. Forty-eight hours later, the Laminaria in each patient were replaced with a rubber balloon (100 mL capacity) that was inflated with 150 to 180 mL of physiologic saline solution, and traction of 800 to 1000 g was applied for an additional 24 hours. During Laminaria application, marked cervical softening was noted in all cases, and weak but definite uterine contractions were initiated in three women. During the balloon application regular uterine contractions were initiated or increased in all patients, with three late second trimester patients aborting within 24 hours. Light and electron microscopic studies of the decidua parietalis obtained during apparent uterine activity and after abortion revealed normal cell morphology. A significant increase in amniotic fluid F prostaglandin was noted after the balloon treatment (P less than .05). The authors conclude that uterine stretching induces uterine activity and a rise of prostaglandin in amniotic fluid without disfunction of decidual cells.

Rupture of the membranes at term, when performed at an appropriate time, is clinically known to induce or accelerate labor. In the present study, the mechanisms of cervical softening and onset and/or enhancement of labor after spontaneous or artificial rupture of the membranes were investigated. The mechanical forces involved in cervical distention at the area where the fetal head is in contact with the cervix were mathematically estimated before and after rupture of the membranes, and the difference between forces under both conditions was calculated. Rupture of the membranes, when occurring either spontaneously or artificially at an appropriate time, has been found to result in an increase of the force of distention in the cervix where it comes into contact with the head. Thus, the increased force against the cervix, as seen by stretching, through the possible increased release of prostaglandins, results in cervical softening and initiation and/or enhancement of labor.
Bleomycin (BLM), a potent anticancer glycopeptide antibiotic, was linked covalently to murine monoclonal anti-HLA IgG1 antibody (H-1) with the use of dextran T-40. As determined spectrophotometrically, the conjugate was composed of 57.5 moles BLM per mole antibody. Of the substituted BLM, 18.4% (10.6 moles BLM per mole antibody) exhibited antimicrobial activity. The BLM-(H-1) conjugate showed stronger cytotoxicity than BLM alone against HLA-bearing cells in cultivation after a 30-min exposure to the drugs. In the same experiment, the conjugate was less toxic than BLM against cells lacking HLA.
The uterus at 48 hours after normal delivery was mechanically stretched by the intra-uterine application of an inflated rubber balloon. Inauguration of regular and marked uterine activity was noted subjectively in all 16 healthy subjects and recorded by an external tocometer. The inaugurated uterine activity was significantly suppressed by the rectal application of 100 mg of indomethacin (p less than 0.01), but was not abolished entirely. The uterine activity ceased gradually with the discontinuation of stretching. These results strongly indicate that the purely stretch-induced uterine contractions are mediated by prostaglandins (PGs) which are released by stretching and/or thereby induced uterine contractions. In this study the possible source of PGs appeared to be by the myometrium itself.
An unspecified region of the uterus of 7 healthy pregnant patients at term, not yet in labor, was mechanically distended by extraamniotic application of rubber bougies. In all cases, significant onset of labor was noted within 20 min to 5 h, cervical softening progressed with time, and delivery was achieved within 21 h. Significant elevation of prostaglandin E (PGE) and F (PGF) levels in amniotic fluid was noted with the progress of cervical dilatation. It appears that the release of PGE and PGF is dependent on uterine stretching and/or the subsequently induced uterine contractions. A possible source of the release of PG in the present study was discussed.
The in vivo antitumor activity of NCS-immune immunoglobulin G (IgG) [Neocarzinostatin (NCS) conjugated with rabbit IgG antibody against a human leukemia cell line (NALL-1)] was evaluated in immunosuppressed newborn Syrian hamsters into which a transplantable human leukemia cell line, BALL-1 was implanted. After intraperitoneal (i.p.) injection of the conjugate, hamsters preinoculated i.p. with BALL-1 cells survived longer than hamsters treated with control solutions (p less than 0.01). The control solutions were NCS, immune IgG, a mixture of NCS and immune IgG, NCS-normal IgG conjugate and physiological saline. There was no change in body weight in the NCS-immune IgG-treated hamsters. The growth of subcutaneously (s.c.) implanted BALL-1 tumors was also inhibited by i.p. administration of NCS-immune IgG; however, the degree of inhibition was not significantly different from that obtained by administration of NCS alone, a mixture of NCS and immune IgG or NCS-normal IgG conjugate. These results indicate that NCS-immune IgG was effective against i.p. BALL-1 tumors, but was less effective against s.c. implanted tumors.
