The glaucomas in pseudophakia.

Abstract

The glaucomas in pseudophakia are not uncommon. Because they can be related to different mechanisms, each case should be evaluated individually. Once glaucoma has been established and medication fails to maintain the intraocular pressure at a safe level, several options are available. As a group, the glaucomas are considered both difficult to manage with medications and also at high risk for failure of filtration surgery. Trabeculectomy adjuvated with anti-metabolites, draining implants, and cyclodestructive procedures are all effective procedures. Although complications and loss of vision as a consequence of glaucoma surgery are not frequent, they were reported by several authors. The likelihood of any given treatment to preserve the quality of life should be discussed with each patient and should be part of the decision-making process. This article reviews studies on the glaucomas in pseudophakia published from October 1994 to September 1995.

Biocompatibility of different barrier membranes in cultures of human CRL 11372 osteoblast-like cells: an immunohistochemical study.

OBJECTIVE: The aim of this study was to evaluate the biocompatibility of two different barrier materials in cultures of human osteoblast-like cells (CRL 11372) in vitro. MATERIAL AND METHODS: Polylactic acid (Epi-Guide; EG) and collagen membrane (Bio-Collagen, BC) were examined. To analyze the effect of materials on cell proliferation, cell numbers and cell viability, cells were cultured on the barrier membranes for 24 and 72 h. Cells plated on culture dishes (CD) served as positive controls. Cell proliferation rate was assessed by the bromodeoxyuridine (BrdU) immunohistochemical technique. The cell numbers of each well were counted. Cell viability was estimated by counting the number of cells, which excluded trypan blue solution. Scanning electron microscopy (SEM) was used to observe the interactions between osteoblastic cells and barrier membranes. RESULTS: The highest number of BrdU-labeled cells were seen on CD after both of the time periods. In comparison with the other two groups, BC showed significantly fewer cells after both time periods. Regarding cell numbers, after 24 and 72 h of incubations CD showed the highest number of cells. The number of viable cells was similar for all the groups. After 72 h for the EG group, SEM view showed flat cells. After 72-h time periods, the BC group revealed a weak adhesion of cells to the barriers. CONCLUSION: The results demonstrate that cells were able to proliferate on these materials and EG promoted the proliferation of human osteoblast like cells. We prefer to use the EG membrane.

4-arylazo-3,5-diamino-1H-pyrazole CDK inhibitors: SAR study, crystal structure in complex with CDK2, selectivity, and cellular effects.

In a routine screening of our small-molecule compound collection we recently identified 4-arylazo-3,5-diamino-1H-pyrazoles as a novel group of ATP antagonists with moderate potency against CDK2-cyclin E. A preliminary SAR study based on 35 analogues suggests ways in which the pharmacophore could be further optimized, for example, via substitutions in the 4-aryl ring. Enzyme kinetics studies with the lead compound and X-ray crystallography of an inhibitor-CDK2 complex demonstrated that its mode of inhibition is competitive. Functional kinase assays confirmed the selectivity toward CDKs, with a preference for CDK9-cyclin T1. The most potent inhibitor, 4-[(3,5-diamino-1H-pyrazol-4-yl)diazenyl]phenol 31b (CAN508), reduced the frequency of S-phase cells of the cancer cell line HT-29 in antiproliferation assays. Further observed cellular effects included decreased phosphorylation of the retinoblastoma protein and the C-terminal domain of RNA polymerase II, inhibition of mRNA synthesis, and induction of the tumor suppressor protein p53, all of which are consistent with inhibition of CDK9.

Synthesis of a technetium-99m labeled tricyclic ganciclovir analog for non-invasive reporter gene expression imaging.

A potential radiopharmaceutical and HSV1-TK substrate, 3-((1,3-dihydroxypropan-2-yloxy)-methyl)-6-(4-(3-((2-mercaptoethyl)(2-(2-mercaptoethyl-amino)-ethyl)amino)propoxy)phenyl)-3H-imidazopurin-9(5H)-one-oxo-technetium(V), was synthesized via a converging approach and its chemical structure was comparatively characterized with a non-radioactive analog. The final radiochemical purity and yield were 97 and 73%, respectively.
