Hypothalamic function in women during the first month post-partum.

Abstract

Plasma LRH-immunoreactive substance in puerperal women was very low on days 5 and 10 post-partum and significantly increased between days 10 and 20 post-partum, but remained at a similar level on day 30 post-partum, which was significantly lower than that in eumenorrhoeic women on day 8 or 9 of the normal cycle. Although LRH administration did not significantly increase serum FSH on day 11 post-partum, daily injections of LRH from days 5 through 10 post-partum caused a significantly increased secretion of FSH on day 11 post-partum, both basally and in response to LRH. No significant difference was observed in serum oestradiol and Prl levels on day 11 post-partum between the two groups of women with and without LRH pre-treatment. It is concluded that the amount of hypothalamic LRH secretion may be small during the first month post-partum, and low basal FSH and unresponsiveness of the pituitary to LRH observed in the early puerperium may result from hypothalamic hypofunction.

Recombinant LH supplementation to recombinant FSH during induced ovarian stimulation in the GnRH-antagonist protocol: a meta-analysis.

This study aims to compare the efficacy of recombinant LH (rLH) supplementation for ovarian stimulation in gonadotrophin-releasing hormone-antagonist protocol for IVF/intracytoplasmic sperm injection cycles. Search strategies included online surveys of databases. The fixed effects model was used for odds ratio (OR) and effect size (weighted mean difference, WMD). Five trials fulfilled the inclusion criteria. When the meta-analysis was carried out, advantages were observed for the LH supplementation protocol with respect to higher serum oestradiol concentrations on the day of human chorionic gonadotrophin administration P < 0.0001; WMD: 514, 95% CI 368, 660) and higher number of mature oocytes (P = 0.0098; WMD: 0.88, 95% CI 0.21, 1.54). However, these differences were not observed in the total amount of recombinant FSH (rFSH) administered, days of stimulation, number of oocyets retrieved, the clinical pregnancy rate per oocyte retrieval, the implantation rate and miscarriage rate. This result demonstrates that the association of rLH with rFSH may prevent any decrease in oestradiol after antagonist administration and that a significantly higher number of mature oocytes was available for laboratory work. Nevertheless, it failed to show any statistically significant difference in clinically significant end-points in IVF (implantation and pregnancy rates). Additional randomized controlled trials are needed to confirm these results further.

Regulation of human estrogen receptor alpha-mediated gene transactivation in Saccharomyces cerevisiae by human coactivator and corepressor proteins.

Human estrogen receptor alpha (ERalpha)-mediated transcription activation was evaluated in the yeast Saccharomyces cerevisiae using both the native ERalpha and a G400V variant. A previous study demonstrated that coexpression of human SRC-1, a potent stimulator of ERalpha function in mammalian cells, potentiated ERalpha-mediated gene expression in yeast over five-fold in an E(2)-dependent manner. In the present study, two additional human coactivator proteins were shown to potentiate ERalpha-mediated gene expression in yeast. SRC2 potentiated transactivation two- to three-fold while SRC3 potentiated transactivation five- to eight-fold. Both human coactivators potentiated both the native ERalpha and the G400V variant in an E(2)-dependent manner. The effect of a human corepressor protein was also evaluated in yeast. Repressor of estrogen receptor activity (REA) did not affect E(2)-induced transactivation by ERalpha (either isoform). However, in a strain that coexpressed human SRC1, REA reduced E(2)-induced transactivation to that observed with ERalpha alone. Furthermore, repression of SRC1 potentiation was specific for the native ERalpha since REA had no effect on SRC1 potentiation of the G400V variant. Additionally, REA repression was specific for SRC1 since potentiation of ERalpha (either isoform) transactivation by SRC2 and SRC3 was unaffected by coexpression of REA. These results support previous observations in mammalian cells that REA does not prevent ERalpha from binding to DNA but does inhibit potentiation of ERalpha-mediated transactivation by SRC1. The results in the present study further characterize REA-mediated repression, and demonstrate the utility of this yeast system for dissecting molecular mechanisms involved in regulating gene transactivation by human ERalpha.

Photodegradation of estrone and 17beta-estradiol in water.

The TiO(2)-assisted photodegradation of two natural female hormones, estrone (E1) and 17beta-estradiol (E2), was investigated in two UV-photo-reactors, followed by solid-phase extraction and analysis by gas chromatography-mass spectrometry (GC-MS). The degradation of E1 and E2 in both reactors followed the pseudo-first-order kinetics. In reactor 1 (150W), 97% of compounds were degraded within 4h of irradiation. Even more rapid degradation was observed in reactor 2 (15W) where 98% of both compounds disappeared within 1h, due to the shorter wavelength of UV-light in reactor 2 (fixed at 253nm) than reactor 1 (238-579nm). The influences of different initial chemical concentrations, pH value, the presence of dissolved organic matter and hydrogen dioxide, and the catalyst concentration on the degradation rate of E1 and E2 in aqueous solutions were investigated. The results show that the extent of photo-induced degradation of E1 and E2 strongly depends on the water constituents in solution. The degradation rate was increased when pH value was increased from 2 to 7.6, beyond which the degradation rate started to decrease. The presence of humic acid enhanced the degradation of E1 and E2 in both reactors as a result of photosensitisation effect of humic acid chromophore. The degradation rate increased with an increase in H(2)O(2) concentration. The degradation rate was also enhanced by increasing catalyst concentration up to 2g/l. The findings therefore suggest that photocatalysis can be a very effective method of rapidly removing certain EDCs from water.
