Comparative study between microscopic agglutination and complement fixation tests in experimental canine leptospirosis.

Abstract

In order to evaluate the antibody response to pathologic leptospira from both serovars icterohaemorrhagiae and canicola, dogs were experimentally inoculated with these leptospiras. Each of the serovar were inoculated in 15 dogs and the antibody response in the acute phase of infected was followed by microscopic agglutination and complement fixation tests. Complement fixing antibodies were detected in both groups at the 4th day post infection and agglutinins appeared in circulation at the 7th day post infection when homologous antigens were used. On the 63rd day post infection with serovar canicola and on 56th day post infection with serovar icterohaemorrhagiae, complement fixing antibodies could no longer be detected but, in both experimental groups, specific agglutinins were presented in significant titers.

General expression of RoTat 1.2 variable antigen type in Trypanosoma evansi isolates from different origin.

The variable surface glycoprotein of Trypanosoma evansi RoTat 1.2 variable antigen type (VAT) is used as an antigen in different antibody detection assays for T. evansi. To obtain more information on the predominant character of RoTat 1.2 and its diagnostic potential in antibody detection tests, we checked its expression in 10 different T. evansi stocks and clones from different parts of the world. Cryostabilates were injected into mice and the trypanosomes of the first peak parasitaemia were screened for the presence of RoTat 1.2 by VAT specific immunofluorescence. To monitor the appearance of RoTat 1.2 specific antibodies during infection, rabbits were infected and serologically tested at different time intervals with VAT specific immune trypanolysis, CATT/T. evansi, LATEX/T. evansi and ELISA/T. evansi. Test results confirm the predominant character of RoTat 1.2.

[Quality of rapid tests for determination of infectious mononucleosis].

BACKGROUND: The Norwegian centre for quality assurance in primary health care, NOKLUS, was established in 1992 to ensure the quality of laboratory analyses performed in primary health care. This article evaluates results from the surveys of infectious mononucleosis. MATERIAL AND METHODS: From 1996 to 2000, five serum panels were sent to participating practices in order to control test-kits designed for serological rapid diagnosis of infectious mononucleosis. 648 practices participated in this external quality assessment in 2000. Target values were determined using the Paul Bunell Davidsohn test. The results obtained for each type of test and variables that might have affected the results were evaluated. RESULTS: Outdated kits or kits close to the date of expiration showed poorer results than the other test-kits. The quality of the results depended mainly on the type of test-kit used and the training level of the persons performing the analyses. INTERPRETATION: The best performing tests in these surveys were Clearview IM (Unipath Limited) and Contrast Mono (Genzyme diagnostics). These two tests are among the three most frequently used tests, out of a total of thirteen, in our surveys.
