SCANCANS--an interactive scanning cell analysis system.

Abstract

This paper describes a computerized, general-purpose, interactive, scanning cell-analysis system developed at the Department of Clinical Cytology of the University Hospital in Uppsala. The system uses a Leitz MPV II scanning photometer, a PDP-8 computer with 24 k of core memory and a Tektronix 4010 graphic display terminal. A command interpreter and overlay structure which makes it simple to add new capabilities to the system is described. Some examples of command strings and the kind of operations the system can perform are included. Finally, the experience gained from using the system and some plans for future extensions are discussed.

[Significance of the automated blood count].

Modern hematology depends on rapid and accurate determination of quantitative and qualitative blood counts. Fully automated analyzers based on the principles of flow cytometry have replaced the traditional blood smear in the routine laboratory. They count the number of red cells, platelets and leukocytes with high precision. In addition, they divide the cells into different populations according to their physical or chemical properties. This population distribution pattern mimics a "differential" blood smear. Normality can be assessed with a high degree of accuracy, rapidly and reproducibly. The qualitative changes frequently provide pointers to the final diagnosis. In case of abnormalities the reports are flagged. A microscopic analysis should be added according to internal laboratory guidelines. It clarifies pathological findings and is essential in the search for specific abnormalities such as malaria. This underlines the need for exchange of information between physician and laboratory.

Fast liquid chromatography for the determination of drugs in plasma and combination with liquid-solid extraction in a fully automated system.

Fast liquid chromatography was applied to the assay of several drugs in plasma. Short columns, 3.3-4 cm long, packed with C18 material, 3 microns particle size, were used. The peaks were little subject to extra-column band-broadening because the investigated drugs were eluted with high capacity factors in order to obtain an adequate separation from plasma components. The main influences on efficiency were the response time of the detector and the solvent composition of the injected sample. Conventional apparatus was used. A fully automated analytical system combining liquid-solid extraction via disposable extraction columns and fast liquid chromatography on a small-dimensioned 3 microns particle size column is described for the assay of drugs in plasma. Automation was accomplished by using the Automatic Sample Preparation with Extraction Columns system.
